Though heterogeneity of cancers is recognized and has been much discussed in recent years, the concept often remains overlooked different routine examinations. Elesclomol induces copper‐dependent ferroptosis in colorectal cancer cells via degradation of ATP7A DOI Creative Commons

Cancer cells reprogram their copper metabolism to adapt adverse microenvironments, such as oxidative stress. The chelator elesclomol has been reported have considerable anticancer efficacy, but the underlying mechanisms remain largely unknown. In this study, we found that elesclomol-mediated overload inhibits colorectal cancer (CRC) both in vitro and vivo. Elesclomol alone promotes degradation of transporter copper-transporting ATPase 1 (ATP7A), which retards proliferation CRC cells. This property distinguishes it from several other chelators. Combinational treatment leads retention within mitochondria due ATP7A loss, leading reactive oxygen species accumulation, turn SLC7A11, thus further enhancing stress consequent ferroptosis effect accounts for robust antitumour activity against CRC, can be reversed by administration antioxidants inhibitors, well overexpression ATP7A. summary, our findings indicate elesclomol-induced chelation targeting regulating ferroptosis.
