The gut microbiota plays an essential role in maintaining immune homeostasis and influencing the landscape within tumor microenvironment. This review aims to elucidate interactions between dynamics, with a focus on colorectal cancer (CRC). spans foundational concepts of immuno-microbial interplay, factors microbiome composition, evidence linking immunotherapy outcomes. Gut modulates anti-cancer immunity through several mechanisms, including enhancement surveillance modulation inflammatory responses. Specific microbial species their metabolic byproducts can significantly influence efficacy immunotherapies. Furthermore, diversity correlates clinical outcomes CRC, suggesting potential as valuable biomarker for predicting response immunotherapy. Conclusions: Understanding relationship responses offers novel therapeutic strategies development. not only influences natural history treatment CRC but also serves critical modulator activity. Further exploration into microbiome's could enhance effectiveness existing treatments guide development new modalities.

Abstract The gut microbiota plays a critical role in maintaining human health, influencing wide range of physiological processes, including immune regulation, metabolism, and neurological function. Recent studies have shown that imbalances composition can contribute to the onset progression various diseases, such as metabolic disorders (e.g., obesity diabetes) neurodegenerative conditions Alzheimer's Parkinson's). These are often accompanied by chronic inflammation dysregulated responses, which closely linked specific forms cell death, pyroptosis ferroptosis. Pathogenic bacteria trigger these death pathways through toxin release, while probiotics been found mitigate effects modulating responses. Despite insights, precise mechanisms influences diseases remain insufficiently understood. This review consolidates recent findings on impact immune‐mediated inflammation‐associated conditions. It also identifies gaps current research explores potential advanced technologies, organ‐on‐chip models microbiome–gut–organ axis, for deepening our understanding. Emerging tools, single‐bacterium omics spatial metabolomics, discussed their promise elucidating microbiota's disease development.

This study investigates the potential of plasma Gasdermin D (GSDMD) as a novel biomarker for early diagnosis and monitoring type 2 diabetes mellitus. We conducted comparative analysis clinical indicators among newly diagnosed patients, those with prediabetes, healthy controls, finding significantly elevated GSDMD levels in group (P < 0.05). Correlation analyses revealed that were positively associated inflammatory markers insulin resistance, while negatively correlating HOMA-β 0.001). Logistic regression identified GSDMD, IL-6, CRP independent risk factors The area under ROC curve was 0.988, indicating superior diagnostic capability compared to traditional like (0.902) IL-6 (0.857). With threshold 17.67 pg/mL, exhibited sensitivity 93.9% specificity 98.0%. These findings suggest not only reflects metabolic abnormalities but also holds promise therapeutic target mitigate responses improve resistance. Further large-scale studies are warranted validate its utility enhance applications.

Pyroptosis is a unique form of programmed cell death characterized by intense inflammation. It involves the activation Gasdermin proteins, which membrane pores, leading to rapid rupture and release inflammatory molecules. Unlike other types death, pyroptosis has distinct mechanisms plays complex role in chronic intestinal diseases, including bowel disease, fibrosis, infectious enteritis, colorectal cancer. This review comprehensively examines how influences disease development progression while exploring therapeutic potential targeting pyroptosis-related pathways. Moreover, interplay between gut microbiota summarized, highlighting its critical pathogenesis disorders. A deeper understanding these diseases may provide valuable insights for future research contribute innovative strategies gastroenterology.

Non-alcoholic fatty liver disease (NAFLD) poses a significant health threat due to its potential progression fibrosis, cirrhosis, and even cancer. Without proper management, NAFLD can lead severe complications significantly impact overall longevity. This study explores the anti-steatosis effects of Nankun Mountain Mao tea (MT) on hepatic lipid accumulation using both in vitro vivo models. In experiments reveal that MT reduces hepatocytes counteracts steatosis induced by palmitic acid oleic acid. investigations high-fat diet (HFD)-fed high-fat, fructose, cholesterol (HFFC)-fed mice demonstrate administration alleviates reducing accumulation, enhancing function, mitigating inflammation. Transcriptomic analyses unveil molecular mechanisms underlying metabolism It turns out inhibits de novo synthesis NF-κB pathway against NAFLD.
