The adult hippocampal neurogenesis plays a vital role in the function of central nervous system (CNS), including memory consolidation, cognitive flexibility, emotional function, and social behavior. deficiency neural stem cells (aNSCs) maintaining quiescence entering cell cycle, self-renewal differentiation capacity is detrimental to functional integrity neurons cognition brain. Histone acetyltransferase (HAT) histone deacetylase (HDAC) have been shown modulate brain functionality are important for embryonic via regulation gene transcription. We showed previously that Trrap, an adapter several HAT complexes, required Sp1 transcriptional control microtubule dynamics neuronal cells. Here, we find Trrap deletion compromises aNSCs mice cultures. acetylation status lysine residues K16, K19, K703 K639 all fail overcome Trrap-deficiency-incurred instability Sp1, indicating scaffold Trrap. Interestingly, deacetylation at greatly increases binding promoter target genes, which antagonizes binding, thereby elevates activity. However, only deacetylated refractory corrects defects Trrap-deleted aNSCs. demonstrate pattern by HATs dictates neurogenesis.

With the development of industrialization in recent years, infrasound has become an important component public noise. To date, diverse studies have revealed negative effects on central nervous system (CNS), especially learning and memory ability. It is widely reported that environmental enrichment (EE) ameliorates deficits different models brain injury. Therefore, present study was designed to determine possible benefits pre-exposure EE preventing functional following exposure their related mechanism. Adult male rats were given enriched or standard housing for 30 days. Following enrichment, exposed 16 Hz, 130 dB 14 days, then ability assessed. Changes neuroinflammation, apoptosis, oxidative stress hippocampus also detected. Our results showed infrasound-induced deficit attenuated significantly pre-exposed rats. Pre-exposure could induce a decrease proinflammatory cytokines increased anti-inflammatory antioxidant properties hippocampus. Moreover, exerted antiapoptosis functions by upregulating B-cell lymphoma/leukemia-2 (Bcl-2) level downregulating P53 In conclusion, suggested neuroprotective when applied before exposure, resulting improved enhancing antioxidant, anti-inflammatory, capacities.

Methadone is used for the treatment of opioid use disorder, including in pregnant patients. Research has established several consequences prenatal misuse, however little work investigated effects methadone exposure (PME) on offspring long-term, despite continued prescription to individuals. The current study aimed identify long-term cognitive impairments arising from PME and assess hippocampal neurogenesis these adult offspring. Pregnant Sprague Dawley rats were injected with or sterile water twice daily gestational day 3-20 left undisturbed as naïve controls. Adult tested one three behavioral tasks pattern separation, spatial learning memory, contextual assigned tissue collection. For assessment neurogenesis, underwent injections bromodeoxyuridine, brains collected at 24hr, 2wks, 4wks immunofluorescent staining. Methadone-exposed females, but not males, showed subtle separation heightened freezing during extinction period fear conditioning task, memory both sexes remained unaffected. Additionally, did alter rate dentate granule cell proliferation significantly reduce number adult-born neuron surviving a mature phenotype females 4wk timepoint. This adds understanding demonstrates female-specific sensitivity consequences. Future needed fully investigate neural disruptions PME, goal better supporting exposed individuals long-term.

Effective preventive measures against postoperative cognitive dysfunction in older adults are urgently needed. In this study, we investigated the effect of electroacupuncture (EA) on anesthesia and surgery-induced decline aged rats by RNA-seq analysis, behavioral testing, Golgi-Cox staining, dendritic spine immunofluorescence assay western blot analysis. EA ameliorated surgery induced-cognitive decline. analysis identified numerous differentially-expressed genes, including 353 upregulated genes 563 downregulated after pretreatment with dysfunction. To examine role CREB EA, injected adeno-associated virus (AAV) into CA1 region hippocampus bilaterally to downregulate transcription factor. improved synaptic plasticity, structurally functionally, activating MAPK/ERK/CREB signaling pathway rats. Together, our findings suggest that protects enhancing hippocampal plasticity.

Abstract As a kind of environmental noise, infrasonic noise has negative effects on various human organs. To date, research shown that infrasound impairs cognitive function, especially the ability for learning and memory. Previously, we demonstrated impaired memory induced by was closely related with glia activation; however, underlying mechanisms remain unclear. Connexin 43 hemichannels (Cx43 HCs), which are mainly expressed in hippocampal astrocytes, activated under pathological conditions, lending support to hypothesis Cx43 HCs might function infrasound. This study revealed blocking or downregulating expression significantly alleviated We also observed exposure led abundant release glutamate ATP through HCs. In addition, depended proinflammatory cytokines. Our finds suggested enhanced astroglial may be involved deficits caused exposure.

Here, we examined whether neurogenic changes evoked by adult‐onset hypothyroidism exhibit sex differences, using female C57BL/6N We assessed impact goitrogen‐induced, on postmitotic survival differentiation hippocampal progenitors Adult‐onset a significant decline neuronal adult‐born within dentate gyrus subfield male, but not female, observed decrease number immature hippocampi hypothyroid mice, whereas goitrogens same treatment paradigms did evoke any change neuron Gene expression analysis euthyroid mice revealed sex‐dependent, differential receptor genes, as well genes linked metabolism transport. Collectively, our findings highlight differences influence neurogenesis, with exhibiting progenitor differentiation. These underscore importance vital variable when considering signaling niche.
