Inflammatory bowel disease (IBD) is a prevalent condition worldwide, characterized by complex etiologies, limited efficacy of clinical drug treatments, and potential adverse effects. In this study, we designed 269 nm selenium nanoparticles with double-cell targeting for ulcerative colitis treatment. Somatostatin (SST) mannooligosaccharide (MOS) were employed to functionalize an Eucommia ulmoides polysaccharide nanoparticle (EUP-SeNP), resulting in the formulation SST/MOS@EUP-SeNP. Nanoparticles engineered target intestinal epithelial cells macrophages through specific cell surface receptors, enabling dual-targeted addition, sodium alginate (SA) microspheres incorporating SST/MOS@EUP-SeNP prepared oral administration, protecting from gastric fluid. The results showed that SA/SST/MOS@EUP-SeNP could preferentially inflamed colon tissue adhere colon, enhance barrier function, regulate level inflammation, antioxidant capacity, composition microbes effectively relieve induced glucan sulfate (DSS). Meanwhile, had excellent biocompatibility both vivo vitro. To some extent, study can provide reference treatment IBD.

Rhein (RH), a natural anthraquinone compound, is considered an effective treatment candidate for ulcerative colitis (UC), whose multiple biological activities contribute to UC, including anti-inflammation, antioxidation, intestinal barrier repair, and microflora regulation. However, the application of RH severely limited by its low water solubility, bioavailability, poor colonic targeting. Although some nanoparticles have been developed oral delivery RH, most them mainly highlighted only one effect drug strategies but above activities. Therefore, polysaccharide-based nanodelivery system, comprising chitosan (CS) fucoidan (FU), with pH/reactive oxygen species (ROS) sensitivity mucosal adhesion, was first used load as comprehensive UC. Briefly, RH-F/C-NPs were prepared using polyelectrolyte self-assembly method; average size 233.1 ± 5.7 nm, encapsulation rate 93.67 1.60%. And it could maintain gastric stability release in colon designed pH/ROS contributed structures. Cellular uptake experiments showed that both NCM 460 cells RAW 264.7 had good RH-F/C-NPs. Importantly, effects vivo experiments, results which significantly reduce DSS-induced inflammation inhibiting TLR4/NF-κB-mediated anti-inflammatory pathway, Nrf2/HO-1-mediated antioxidant In addition, pharmacokinetic studies shown F/C-NPs increase plasma concentration accumulation extent. short, this study develop nanosystem dual "one stone four birds" therapeutic on

This review aims to identify in vivo studies investigating the potential of plant substances and their natural molecules managing inflammatory bowel disease (IBD). Specifically, objective is examine impact these on interleukins other key signaling markers. Relevant articles published up December 2022 were identified through a search PubMed, Scopus, Web Science, Embase databases. The used keywords including "inflammatory disease", "medicinal plants", "natural molecules", "anti-inflammatory", "ulcerative colitis", 1,878 potentially relevant articles, which 89 included this after completion selection process. study provides preclinical data products (NPs) that can treat IBD, ulcerative colitis. main actions NPs relate effects nuclear factor kappa B (NF-κB), Janus kinase (JAK)/signal transducer activator transcription (STAT) pathway, regulation T helper 17/regulatory cells balance, oxidative stress. ability inhibit intestinal inflammation appears be dependent lowering levels pro-inflammatory cytokines tumor necrosis factor-alpha (TNF-α), interleukin (IL)-1β, IL-17, via Jun N-terminal (JNK)1, NF-κβ-p65, STAT3 pathways. In addition, shown reduce stress severity colitis, as well increase activity antioxidant enzymes. These suggest represent promising treatment for have greater efficacy safety than current treatments.

Coumarin, a naturally occurring compound found in various plants, has rich history of use traditional medicine. Recent research highlighted its anti-inflammatory properties, positioning it as promising candidate for treating inflammatory disorders such rheumatoid arthritis, asthma, and bowel disease. This narrative review aims to comprehensively summarize the current knowledge regarding coumarin's pharmacological effects alleviating conditions by analyzing preclinical clinical studies. The focuses on elucidating mechanisms through which coumarin exerts effects, including antioxidant activity, inhibiting pro-inflammatory cytokine production, modulation immune cell functions. Additionally, paper addresses potential limitations using coumarin, concerns about toxicity at high doses or with prolonged use. Before widespread application, further investigation is needed fully understand benefits risks.

Inflammatory bowel disease (IBD) is a chronic autoimmune disorder stemming from unrestrained immune activation and subsequent destruction of colon tissue. Genetic susceptibility, microbiota remodeling, environmental cues are involved in IBD pathogenesis. Up to now, there limited treatment options for IBD, so better therapies eagerly needed. The therapeutic effects naturally occurring compounds have been extensively investigated, among which quercetin becomes an attractive candidate owing its unique biochemical properties. To facilitate the clinical translation quercetin, we aimed get comprehensive understanding cellular molecular mechanisms underlying anti-IBD role quercetin. We summarized that exerts effect through consolidating intestinal mucosal barrier, enhancing diversity colonic microbiota, restoring local homeostasis, restraining oxidative stress response. also delineated on gut microbiome discussed potential side administration. Besides, could serve as prodrug, bioavailability improved chemical modifications or utilization effective drug delivery systems. Altogether, these lines evidence hint feasibility compound treatment.

Ulcerative colitis (UC) is associated with brain neurotransmitter disorders and intestinal dysbiosis. Bacillus amyloliquefaciens fmb50 produces the lipopeptide surfactin, which has a wide range of biological activities. However, effects surfactin on DSS-induced have not been reported. In present study, oral significantly ameliorated in mouse model reduced depression-like behavior, such as slowed walking speed, shortened movement distance open field test, weakened exploration ability light–dark shuttle test. Surfactin noticeably improved gut microbial dysbiosis, barrier dysfunction colon, blood–brain brain. Furthermore, colon levels occludin were upregulated by 68.51%, ZO-1 77.81% 36.42%, respectively. supplementation also inhibited inflammatory responses inactivating tumor necrosis factor-α (TNF-α), nuclear factor kappa-B (NF-κB), NLRP3 signaling pathways Thus, we believe that behavioral upregulating 5-hydroxytryptamine (5-HT), 5-hydroxyindoleacetic acid (5-HIAA), norepinephrine (NE), brain-derived neurotrophic (BDNF), suppressing responses, improving dysfunction. abundances microbes are related to colitis, especially targeting facultative anaerobes phylum Proteobacteria, it increased abundance beneficial bacteria Lactobacillus unidentified Prevotella. Combined its nontoxic nature observed this long-term study mice, might be promising intervention strategy for preventing acting microbiota-gut–brain axis.
