Abstract Background: Cardiac Myxoma (CM) is a primary tumor of heart. Its origins, why the occurrence cardiac tumors extremely rare and how it may be related to limited regenerative potential, these aspects are not yet entirely known. This study investigates key genes/TFs signaling pathways advance our understanding important questions. Methods: Databases including PubMed, MEDLINE, Google Scholar were searched for published articles without any date restrictions, involving myxoma, genes/TFs/signaling their roles in cardiogenesis, proliferation, differentiation, interactions tumorigenesis, with focus on cardiomyocytes. Sodium-glucose cotransporters 2 (SGLT2) are high-capacity, low-affinity transporters, expressed mainly in the early portion of proximal renal tube, mediating up to 90% glucose uptake, while SGLT1 receptors found small intestine, facilitating absorption. SGLT2 inhibitors (SGLT2i) originally emerged as agents for treatment type diabetes mellitus; however, they soon demonstrated remarkable cardio- and renoprotective actions that led their licensed use heart failure chronic kidney disease, regardless diabetic status. ABSTRACT Cardiac hypertrophy, a maladaptive response to chronic stress, progresses heart failure through mechanisms requiring deeper exploration. While forkhead helix transcription factor P3 (FoxP3) is well‐known as key regulator in CD4 + T cells, its role cardiomyocytes remains unclear. Here, by using isoproterenol (ISO)‐induced cardiac hypertrophy models (40 mg/kg daily for vivo study and 10 μmol/L vitro study), we revealed the protective of FoxP3 hypertrophy. Though modulating expression siRNA or plasmid cardiomyocytes, found that knockdown exacerbated ISO‐induced hypertrophic responses, while overexpression attenuated effects. The function cardiomyocytic was further confirmed infection adeno‐associated virus. Mechanically, decreased nuclear activated cells c3 (NFATc3), hypertrophy‐related genes, suppress including atrial natriuretic peptide, brain peptide β‐myosin heavy chain (β‐MHC), thus ameliorate responses.
