Abstract Incomplete autophagy is an impaired self‐eating process of intracellular macromolecules and organelles in which accumulated autophagosomes do not fuse with lysosomes for degradation, resulting the blockage autophagic flux. In this review, we summarized literature over past decade describing incomplete autophagy, found that different from double‐edged sword effect general on promoting cell survival or death, plays a crucial role disrupting cellular homeostasis, promotes only death. What matters closely relevant to pathogenesis progression various human diseases, which, meanwhile, intimately linking pharmacologic toxicologic effects several compounds. Here, comprehensively reviewed latest progress molecular mechanisms signaling pathways. Moreover, implications pharmacotherapy are also discussed, has great relevance our understanding distinctive physiology disease. Consequently, targeting may contribute development novel generation therapeutic agents diverse diseases.

Abstract Mitophagy is a vital process that controls mitochondria quality, dysregulation of which can promote cancer. Oncoprotein mucin 1 (MUC1) targets to attenuate drug-induced apoptosis. However, little known about whether and how MUC1 contributes mitochondrial homeostasis in cancer cells. We identified novel role promoting mitophagy. Increased mitophagy coupled with the translocation mitochondria, where interacts induces degradation ATPase family AAA domain-containing 3A (ATAD3A), resulting protection PTEN-induced kinase (Pink1) from ATAD3A-mediated cleavage. Interestingly, MUC1-induced associated increased oncogenicity Similarly, inhibition significantly suppresses cell activity vitro vivo. Consistently, ATAD3A protein levels present an inverse relationship tumor tissues breast patients. Our data validate MUC1/ATAD3A/Pink1 axis-mediated constitutes mechanism for maintaining malignancy cells, providing therapeutic approach MUC1-positive cancers.

Skeletal muscle is one of the largest organs in body and essential for maintaining quality life. Loss skeletal mass function can lead to a range adverse consequences. The gut microbiota interact with by regulating variety processes that affect host physiology, including inflammatory immunity, protein anabolism, energy, lipids, neuromuscular connectivity, oxidative stress, mitochondrial function, endocrine insulin resistance. It proposed plays role direction work. Even though notion microbiota-muscle axis (gut-muscle axis) has been postulated, its causal link still unknown. impact on described detail this review.

Autophagy is an evolutionarily conserved and tightly regulated process that plays important role in maintaining cellular homeostasis. It involves regulation of various genes function to degrade unnecessary or dysfunctional components, recycle metabolic substrates. modulated by many factors, such as nutritional status, energy level, hypoxic conditions, endoplasmic reticulum stress, hormonal stimulation drugs, these factors can regulate autophagy both upstream downstream the pathway. In cancer, acts a double-edged sword depending on tissue type stage tumorigenesis. On one hand, promotes tumor progression advanced stages stimulating growth. other inhibits development early enhancing its suppressor activity. Moreover, drives resistance anticancer therapy, even though some types, activation induces lethal effects cancer cells. this review, we summarize biological mechanisms dual cancer. addition, report current understanding types with markedly high incidence and/or lethality, existing therapeutic strategies targeting for treatment

Wilms’ tumor 1-associating protein (WTAP) is required for N6-methyladenosine (m6A) RNA methylation modifications, which regulate biological processes such as splicing, cell proliferation, cycle, and embryonic development. m6A the predominant form of mRNA modification in eukaryotes. WTAP exerts by binding to methyltransferase-like 3 (METTL3) nucleus METTL3-methyltransferase-like 14 (METTL14)-WTAP (MMW) complex, a core component methyltransferase complex (MTC), localizing nuclear patches. Studies have demonstrated that plays critical role various cancers, both dependent independent its methyltransferases. Here, we describe recent findings on structural features WTAP, mechanisms regulates functions, molecular functions cancers. Betulinic acid (BA), a pentacyclic triterpenoid isolated from tree bark, exhibits antitumor effects against solid malignancies and triggers autophagy and/or apoptosis in human cancer cells. Nonetheless, the relationship between potential modulatory actions of BA on autophagy-dependent bladder cell death remain unclear. Although previous studies have reported that saponins (ginsenosides, the major active and most representative ingredients in Panax ginseng C.A. Meyer) exerted a good ameliorative effect on cisplatin (CP)-induced acute kidney injury animal models, little attention has been paid to large number of polysaccharides isolated purified from ginseng. This work aimed investigate protective possible molecular mechanism polysaccharide (WGP) CP-induced toxicology mice. The results biomarker analysis including serum creatinine (CRE) blood urea nitrogen (BUN) confirmed WGP at 200 400 mg/kg renal-toxicology. We found reduces apoptosis cells by inhibiting endoplasmic reticulum (ER) stress caused CP, which is manifested increased phosphorylation PERK. In addition, apoptosis-associated with caspase 3 activation renal induced CP was inhibited after administration WGP, levels PI3K AKT were also reduced significantly. demonstrated exposure unfolded protein response signaling pathway PERK-eIF2α-ATF4 axis significantly activated, eIF2α expression ATF4 CHOP.
