Cadmium (Cd) and high molybdenum (Mo) are injurious to the body. Previous research has substantiated that Cd Mo exposure caused testicular injury of ducks, but concrete mechanism is not fully clarified. To further survey toxicity co-exposure in testis, 40 healthy 8-day-old Shaoxing ducks (Anas platyrhyncha) were stochasticly distributed four groups raised with basic diet embracing (4 mg/kg Cd) or (100 Mo) both. At 16th week, testis tissues gathered. The characteristic ultrastructural changes related apoptosis ferroptosis observed both groups. Besides, repressed nuclear factor erythroid 2-related 2(Nrf2) pathway via decreasing Nrf2, Heme oxygenase-1 (HO-1), NAD(P)H quinone oxidoreductase 1 (NQO1), Glutamate-cysteine ligase catalytic subunit (GCLC) modifier (GCLM) mRNA expression Nrf2 protein expression, then stimulated by elevating Bcl-2 antagonist/killer-1 (Bak-1), Bcl-2-associated X-protein (Bax), Cytochrome complex (Cyt-C), caspase-3 cleaved-caspase-3 rate, as well reducing B-cell lymphoma-2 (Bcl-2) ratio Bax, triggered upregulating Acyl-CoA Synthetase Long Chain Family Member 4 (ACSL4), transferrin receptor (TFR1) Prostaglandin-Endoperoxide Synthase 2 (PTGS2) levels, downregulating ferritin heavy chain (FTH1), light (FTL1), ferroportin (FPN1), solute carrier family 7 member 11 (SCL7A11) glutathione peroxidase (GPX4) levels. most obvious these indexes co-treated group. Altogether, results announced evoked inhibiting exacerbated variations.

Abstract Urbanisation and industrialisation are on the rise all over world. Environmental contaminants such as potentially toxic elements (PTEs) directly linked with both phenomena. Two PTEs that raise greatest concern arsenic (As) lead (Pb) soil drinking water contaminants, whether they naturally occurring or consequence of human activities. Both potential carcinogens. This paper reviews mechanisms by which As Pb impair metabolic processes cause genetic damage in humans. Despite efforts to ban limit their use, due high persistence continue pose a risk health, justifies need for further toxicological research.

Abstract Cadmium (Cd) is a global pollutant, and its accumulation in the liver causes oxidative stress, inflammation, insulin resistance (IR), metabolic complications. This study investigated whether curcumin treatment could alleviate hepatic IR Wistar rats exposed to sub-chronic cadmium explored underlying molecular pathways. Male were divided into control group (standard normocaloric diet + cadmium-free water) drinking water with 32.5 ppm CdCl 2 ) for 30 days. Oral glucose tolerance, response, assessed using mathematical models. Liver tissue was analyzed markers of key regulatory pathways, including NF-κB, Nrf2, MAPKs (JNK p38), IRS1-Akt pathway. We established an effective dose 250 mg/kg 5 days orally. Results demonstrated that after exposure, accumulated liver, inducing inflammatory state. characterized by increased expression JNK, p38, along diminished Nrf2 expression, IR, hyperglycemia, hyperinsulinemia. Curcumin effectively alleviated these disorders restoring balance between NF-κB modulating MAPK pathway, and, consequently, improving balance. In conclusion, this suggests induces through imbalance signaling appears improve thereby ameliorating IR.

Epidemiology studies have indicated that environmental cadmium exposure, even at low levels, will result in chronic accumulation the kidney with profound adverse consequences and diabetic population is more susceptible. However, underlying mechanisms are yet not fully understood. In present study, we applied an animal model to study exposure-induced renal injury performed whole transcriptome profiling studies. Repetitive CdCl2 exposure resulted remarkable injuries animals. The ob/ob mice manifested increased severity of compared wild type C57BL/6 J littermate controls. RNA-Seq data showed treatment induced dramatic gene expression changes a dose-dependent manner. Among differentially expressed genes include apoptosis hallmark which significantly demarcated effects. Pathway enrichment network analyses revealed biological oxidation (mainly glucuronidation) as one major stress responses by treatment. We next implemented deep learning algorithm conjunction cloud computing discovered signature can predict degree provided, for first time, comprehensive mechanistic understanding cadmium-induced nephrotoxicity normal populations genome level.
