Abstract Non-coding RNA has aroused great research interest recently, they play a wide range of biological functions, such as regulating cell cycle, proliferation, and intracellular substance metabolism. Piwi-interacting RNAs (piRNAs) are emerging small non-coding that 24–31 nucleotides in length. Previous studies on piRNAs were mainly limited to evaluating the binding PIWI protein family role. However, recent have shed more lights piRNA functions; aberrant unique roles many human diseases, including diverse lethal cancers. Therefore, understanding mechanism expression specific functional diseases is crucial for developing its clinical applications. Presently, focuses their cancer-specific functions but lacks investigation expressions epigenetic modifications. This review discusses piRNA’s biogenesis progress piRNA/PIWI complexes diseases.

Preventing local tumor recurrence while promoting bone tissue regeneration is an urgent need for osteosarcoma treatment. However, the therapeutic efficacy of traditional photosensitizers limited, and they lack ability to regenerate bone. Here, a piezo-photo nanoheterostructure developed based on ultrasmall bismuth/strontium titanate nanocubes (denoted as Bi/SrTiO3), which achieve piezoelectric field-driven fast charge separation coupling with surface plasmon resonance efficiently generate reactive oxygen species. These hybrid nanotherapeutics are integrated into injectable biopolymer hydrogels, exhibit outstanding anticancer effects under combined irradiation NIR ultrasound. In vivo studies using patient-derived xenograft models tibial demonstrate that hydrogels suppression rates 98.6 % 67.6 in respective models. Furthermore, hydrogel had good filling retention capabilities defect region, exerted repair by polarizing conveying electrical stimuli cells mild ultrasound radiation. This study provides comprehensive clinically feasible strategy overall treatment osteosarcoma.

In the last decades, treatment of primary and secondary bone tumors has faced a slow-down in its development, being mainly based on chemotherapy, radiotherapy, surgical interventions. However, these conventional therapeutic strategies present series disadvantages (e.g., multidrug resistance, tumor recurrence, severe side effects, formation large defects), which limit their application efficacy. recent years, procedures were combined with several adjuvant therapies, different degrees success. To overcome drawbacks current therapies improve outcomes, other started investigated, like carrier-mediated drug delivery, substitutes for repairing defects, multifunctional scaffolds tissue regeneration antitumor properties. Thus, this paper aims to types approaches, further focusing advances new alternatives.

Osteosarcoma (OS) is the most common primary malignant bone sarcoma mainly affecting adolescents and young adults, which often progresses to pulmonary metastasis leads death of OS patients. characterized as a highly heterogeneous cancer type underlying pathologic mechanisms triggering tumor progress are incompletely recognized. Surgery combined with neoadjuvant postoperative chemotherapy has elevated 5-year survival over 70% for patients localized tumors, opposed only 20% recurrence and/or metastasis. Therefore, novel therapeutic strategies needed overcome drawbacks conventional treatments. Immunotherapy gaining momentum treatment an increasing number FDA-approved therapies malignancies resistant therapies. Here, we review microenvironment appraise promising immunotherapies available in management OS.

Osteosarcoma (OS) is the most common malignant bone tumor in children. Despite efforts to develop and implement new therapies, patient outcomes have not measurably improved since 1980s. Metastasis continues be main source of mortality, with 30% cases developing metastatic disease within 5 years diagnosis. Research models are critical advancement cancer research include a variety species. For example, xenograft patient-derived (PDX) mouse provide opportunities study human cells vivo while transgenic offered significant insight into molecular mechanisms underlying OS development. A growing recognition naturally occurring cancers companion species has led insights how veterinary patients can contribute studies biology drug The canine cases, including use diagnostic tissue archives clinical trials, offers potential mechanism further research. Advancement field requires continued development appropriate animal models. In this review, described focus on tumor-bearing pet dog as parallel complementary OS.
