Perfluoroalkyl substances (PFAS) are environmental contaminants present in a wide range of consumer products and frequently detected drinking water. They have been linked to adverse reproductive health outcomes women, but there is limited human evidence on the association PFAS exposure with endometriosis. Pregnane X receptor (PXR) is a heterologous biosensor that involved in the metabolic pathway of environmental pollutants, regulating transcription genes biotransformation. There are significant differences selectivity and specificity organic pollutants (OPs) toward polar bear PXR (pbPXR) human (hPXR), but detailed dynamical characteristics their interactions unclear. Homology Modeling, molecular docking, dynamics simulation, free energy calculation were used to analyze recognition pbPXR hPXR by three OPs: BPA, chlordane toxaphene. Comparing interaction patterns along with binding these OPs revealed although interact similar OPs, have different effects on internal hPXR. This results alterations key residues near Leu209, Met243, Phe288, Met323, His407 thereby influencing energy. Non-polar interactions, especially van der Waals found be dominating factors interacting PXRs. The region surrounding facilitates hydrophobic contacts PXR, which crucial for selective activation PXRs species OPs. These findings guidance understanding impacts endocrine disruptors organisms.
