Combination therapies demand co-delivery platforms with efficient entrapment of distinct payloads and specific delivery to cells possibly organelles. Herein, we introduce the combination two therapeutic modalities, gene photodynamic therapy, in a purely peptidic platform. The simultaneous formation cargo loading multi-micellar platform is governed by self-assembly at nanoscale. architecture nanocarrier positive charge its constituent micelles offer controlled dual capacity locations for hydrophobic photosensitizer (PS) negatively charged antisense oligonucleotides (ASOs). Moreover, nuclear localization signal (NLS) sequence built-in peptide targets PS + ASO-loaded nanocarriers nucleus. Breast cancer treated demonstrated photo-triggered enhancement radical oxygen species (ROS) associated increased cell death. Besides, ASO resulted up 90 % knockdown Bcl-2, an inhibitor apoptosis that overexpressed more than half all human cancers. Simultaneous elicited synergistic extent could not be reached singly loaded or free form drugs. Both, location compounds prevents them from interfering each other, highly cellular support great potential this versatile therapy.

Atherosclerosis is the build-up of fatty deposits in arteries. main underlying cause cardiovascular diseases and leading global morbidity mortality. Current pharmaceutical treatment options are unable to effectively treat plaque later stages disease. Instead, they aimed at resolving risk factors. Nanomaterials nanoparticle-mediated therapies have become increasingly popular for atherosclerosis due their targeted controlled release therapeutics. In this review, we discuss different types therapeutics used disease focus on nanomaterial strategies employed delivery these drugs, enabling effective efficient resolution atherosclerotic plaque. The ideal strategy each drug type (e.g. statins, nucleic acids, small molecule peptides) will be comprehensively discussed.

Improving the efficacy of chemotherapy remains a key challenge in cancer treatment, considering low bioavailability, high cytotoxicity, and undesirable side effects some clinical drugs. Targeted delivery sustained release therapeutic drugs to cells can reduce whole-body cytotoxicity agent deliver safe localized treatment patient. There is growing interest herbal drugs, such as curcumin, which highly noted promising anti-tumor drug, its wide range bioactivities properties against various tumors. Conversely, curcumin limited because poor oral water solubility, instability gastrointestinal fluids, unsuitable pH stability. Drug-delivery colloid vehicles like liposomes nanoparticles combined with microbubbles ultrasound-mediated are currently being explored effective modes cases. This study aimed synthesize (CLs) optimize high-frequency ultrasound uptake by human breast cell line (HCC 1954) through vitro studies culture viability cytotoxicity. CLs were effectively prepared particles sized at 81 ± 2 nm, demonstrating stability controlled under exposure. In using HCC1954 cells, combination CLs, ultrasound, Definity significantly improved curcumin's effects, particularly specific conditions: 15 s continuous 0.12 W/cm

Abstract Rheumatoid arthritis (RA) is an autoimmune disease with multifactorial etiology and intricate pathogenesis. In RA, repeated monotherapy frequently associated inadequate efficacy, drug resistance, severe side effects. Therefore, a shift has occurred in clinical practice toward combination therapy. However, conventional therapy encounters several hindrances, including low selectivity to arthritic joints, short half-lives, varying pharmacokinetics among coupled drugs. Emerging nanotechnology offers incomparable opportunity for developing advanced against RA. First, it allows co-delivering multiple drugs augmented physicochemical properties, targeted delivery capabilities, controlled release profiles. Second, enables therapeutic nanomaterials development, thereby expanding regimens include multifunctional nanomedicines. Lastly, facilitates the construction of all-in-one nanoplatforms assembled modalities, such as phototherapy, sonodynamic therapy, imaging. Thus, promising solution current bottleneck both RA treatment diagnosis. This review summarizes rationale, advantages, recent advances nano-empowered It also discusses safety considerations, drug–drug interactions, potential translation. Additionally, provides design tips outlook on future developments The objective this achieve comprehensive understanding mechanisms underlying unlock maximum nanotechnology, facilitating smooth transition research findings from laboratory practice.
