Abstract The aftermath of traumatization lives on in the neural and epigenetic traces creating a momentum affliction psychological social realm. Using randomised controlled parallel group design, we examined methylome-wide saliva samples 84 female former child soldiers from Eastern DR Congo before six months after Narrative Exposure Therapy. Treatment predicted differentially methylated positions (DMPs) related to ALCAM , RIPOR2 AFAP1 MOCOS . In addition, treatment associations overlapped at gene level with baseline clinical outcomes. A dose-dependent effect of Adverse Childhood Experiences (ACE) on the course and severity psychiatric disorders has been frequently reported. Adverse childhood experiences (ACEs) increase the likelihood of reduced physical and psychological health in adulthood. Though understanding management traumatic is growing, empirical exploration ACEs clinical outcomes remains under-represented under-explored. The wars in the Democratic Republic of Congo have left indelible marks on mental health and functioning Congolese civilians that sought refuge Uganda. Abstract Epigenetic processes allow plasticity in gene regulation response to significant environmental events. Accumulating evidence suggests that effective psychotherapy is accompanied by epigenetic changes, rendering DNA methylation a potential biomarker of therapy success. We investigated associations symptom development changes 38 CpG sites spanning over three times measurement using repeated measures correlation. After accounting for multiple comparisons, site cg25535999 remained negatively associated symptoms. Our suggest different profile compared non-responders underscore signaling trauma-focused therapy.
