Nervous systems function as ensembles of neurons communicating via synaptic connections, and a functional understanding nervous requires extensive knowledge their connectomes. In companion paper (Takemura et al., 2023), we describe the acquisition complete fruit fly nerve cord connectome, first for an animal that can walk or fly. Here, to efficiently navigate appreciate biological significance this categorise name nearly all systematically link them experimental literature. Neural circuit mapping is generating datasets of tens thousands labeled neurons. New computational tools are needed to search and organize these data. We present NBLAST, a sensitive rapid algorithm, for measuring pairwise neuronal similarity. NBLAST considers both position local geometry, decomposing neurons into short segments; matched segments scored using probabilistic scoring matrix defined by statistics matches non-matches. validated on published dataset 16,129 single Drosophila can distinguish types down the finest level (single identified neurons) without priori information. Cluster analysis extensively studied classes new unreported topographical features. Fully automated clustering organized validation 1,052 clusters, many which map onto previously described types. supports additional query types, including searching against transgene expression patterns. How brains are hardwired to produce aggressive behavior, and how aggression circuits related those that mediate courtship, is not well understood. A large-scale screen for aggression-promoting neurons in Drosophila identified several independent hits enhanced both inter-male courtship. Genetic intersections revealed 8-10 P1 interneurons, previously thought exclusively control male were sufficient promote fighting. Optogenetic experiments indicated activation could at a threshold below required wing extension. the absence of extension triggered persistent via an internal state endure minutes. High-frequency promoted suppressed during photostimulation, whereas resumed was inhibited following photostimulation offset. Abstract Oestradiol establishes neural sex differences in many vertebrates 1–3 and modulates mood, behaviour energy balance adulthood 4–8 . Many motor control systems generate multiple movements using a common set of muscles. How are premotor circuits able to flexibly diverse movement patterns? Here, we characterize the neuronal that drive distinct courtship songs Drosophila melanogaster. Each taste modality is generally encoded by a single, molecularly defined, population of sensory cells. However, salt stimulates multiple pathways in mammals and insects, suggesting more complex code for taste. Here, we examine coding Drosophila. After creating comprehensive molecular map comprised five discrete neuron classes across the fly labellum, find that four are activated salt: two exhibiting characteristics ‘low salt’ cells, ‘high classes. Behaviorally, low attraction depends primarily on ‘sweet’ neurons, with additional input from neurons expressing ionotropic receptor IR94e. High avoidance mediated ‘bitter’ glutamatergic Ppk23. Interestingly, impact these prior consumption. These results support model flies combinatorially integrates inputs cell types to afford robust flexible behaviors.
