In Drosophila, a dedicated olfactory channel senses male pheromone, cis-vaccenyl acetate (cVA), promoting female courtship while repelling males. Here, we show that separate cVA-processing streams extract qualitative and positional information. cVA sensory neurons respond to concentration differences in 5-mm range around male. Second-order projection encode the angular position of by detecting inter-antennal concentration, which are amplified through contralateral inhibition. At third circuit layer, identify 47 cell types with diverse input-output connectivity. One population responds tonically flies, second is tuned looming, integrates taste coincidentally promote mating. The separation features resembles mammalian what where visual streams; together multisensory integration, this enables behavioral responses appropriate specific ethological contexts.

It remains unclear how memory engrams are altered by experience, such as new learning, to cause forgetting. Here, we report that short-term aversive in Drosophila is encoded and retrieved from the mushroom body output neuron MBOn-γ2α′1. Pairing an odor with electric shock creates a robust depression calcium response of MBOn-γ2α′1 increases avoidance paired odor. Electric after which activates cognate dopamine DAn-γ2α′1, restores properties causes behavioral Conditioning second responses previously learned while depressing newly odor, showing learning forgetting can occur simultaneously. Moreover, optogenetic activation DAn-γ2α′1 sufficient for bidirectional modulation properties. Thus, single DAn drive both bidirectionally modulating cellular trace.

ABSTRACT Recent developments in machine vision methods for automatic, quantitative analysis of social behavior have immensely improved both the scale and level resolution with which we can dissect interactions between members same species. In this paper, review these methods, a particular focus on how biologists apply them to their own work. We discuss several components vision-based analyses: record high-quality video automated analyses, video-based tracking algorithms estimating positions interacting animals, learning recognizing patterns interactions. These are extremely general applicability, subset successful applications biological questions model systems very different types behaviors.

Aggressive social interactions are used to compete for limited resources and regulated by complex sensory cues the organism's internal state. While both sexes exhibit aggression, its neuronal underpinnings understudied in females. Here, we identify a population of sexually dimorphic aIPg neurons adult Drosophila melanogaster central brain whose optogenetic activation increased, genetic inactivation reduced, female aggression. Analysis GAL4 lines identified an unbiased screen increased chasing behavior revealed involvement another neuron, pC1d, implicated pC1d as core nodes regulating Connectomic analysis demonstrated that interconnected suggest may exert part their effect gating flow visual information descending neurons. Our work reveals important regulatory components circuitry underlies aggressive provides tools manipulation.
