Abstract Social recognition memory (SRM) is a key determinant of social interactions. While the cerebellum emerges as an important region for behavior, how cerebellar activity affects functions remains unclear. We selectively increased excitability molecular layer interneurons (MLIs) to suppress Purkinje cell firing in mouse vermis. Chemogenetic perturbation MLIs impaired SRM without affecting sociability, anxiety levels, motor coordination or object recognition. Optogenetic interference during distinct phases test revealed engagement retrieval, but not encoding, information. c-Fos mapping after showed that manipulation decreased brain-wide interregional correlations and altered network structure from medial prefrontal cortex hippocampus-centered amygdala-centered modules. Anatomical tracing demonstrated hierarchical projections central brain integrating amygdalar connections. Our findings suggest organizes neural matrix necessary SRM.

Ventral hippocampus (vHIP) and medial prefrontal cortex (mPFC) are both critical regions for social behaviors. However, how their interactions affect behavior is not well understood. By viral tracing, optogenetics, chemogenetics, fiber photometry, we demonstrated that inhibition of vHIP or direct projections from to mPFC impaired memory expression. Via rabies retrograde found all three major GABAergic neurons in received inputs vHIP. Activation parvalbumin positive (PV+) but somatostatin (SST+) can rescue the impairment caused by inhibition. Furthermore, photometry results behaviors preferentially recruited PV+ hippocampal disrupted activity during interactions. These revealed a new mechanism regulate complementarity with existing neural circuitry mechanism.

Neuroscientists have adopted several model organisms study in laboratory setting, including fish, reptiles, rodents primates.
