Adverse childhood experiences (ACEs) are common in people at clinical high-risk for psychosis (CHR), however, the relationship between ACEs and long-term outcomes is still unclear. This study examined associations CHR individuals. 344 individuals 67 healthy controls (HC) were assessed using Childhood Trauma Questionnaire (CTQ), Bullying Experience of Care Abuse (CECA). followed up to 5 years. Remission from state, transition (both defined with Comprehensive Assessment an At Risk Mental State), level functioning (assessed Global Functioning) assessed. Stepwise multilevel logistic regression models used investigate outcomes. significantly more prevalent than HC. Abstract The major anxiety disorders (ANX; including generalized disorder, panic and phobias ) are highly prevalent, often onset early, persist throughout life, cause substantial global disability. Although distinct in their clinical presentations, they likely represent differential expressions of a dysregulated threat-response system. Here we present genome-wide association meta-analysis comprising 122,341 European ancestry ANX cases 729,881 controls. We identified 58 independent significant risk variants 66 genes with robust biological support. In an sample 1,175,012 self-report 1,956,379 controls, 51 the associated were replicated. As predicted by twin studies, found genetic correlation between depression, neuroticism, other internalizing phenotypes. Follow-up analyses demonstrated enrichment all brain regions highlighted GABAergic signaling as one potential mechanism underlying risk. These results advance our understanding architecture prioritize for functional follow-up studies.

Association between stressful life events and depression, anxiety, and quality of life among urban adolescents and young adults in Latin America DOI Creative Commons

Background Latin American youth have a high prevalence of mental health disorders and face major socioeconomic public safety problems. This study assesses the association between stressful life events (SLEs) depression, anxiety, quality among adolescents young adults from deprived neighbourhoods America. Methods The sample consisted 2,402 participants, (15–16 years) (20–24 years), Bogotá (Colombia), Buenos Aires (Argentina), Lima (Peru), assessed in 2021–2022 recruited education community settings social media. We evaluated most frequent recent distant SLEs (occurred previous year more than ago, respectively), relationship severity depression (PHQ-8), anxiety (GAD-7), (MANSA), we tested for differences by gender age group. Results common were related to issues financial distress. Accidents school suspensions men, sexual harassment bullying women. Every additional reported SLE increased odds mild, moderate, severe (18, 17, 25%, respectively) (10, 21%, symptoms. Similar trends found with (8, 9, 11% symptoms, (9, 11, 12%, respectively). Furthermore, higher number associated lower ( β = −0.05, p < 0.001, 95% CI [−0.06, −0.04] −0.04, [−0.05, −0.03], distress seems stronger one SLEs, may impact on adolescents’ compared adults. Conclusion Both are life. Policies programmes should aim enhance safety, as well improve individual, family, protective factors that could mitigate effect youth.

Childhood adversity is a strong predictor of developing psychopathological conditions. Multiple theories on the mechanisms underlying this association have been suggested which, however, differ in operationalization ‘exposure.’ Altered (threat) learning represent central by which environmental inputs shape emotional and cognitive processes ultimately behavior. 1402 healthy participants underwent fear conditioning paradigm (acquisition training, generalization), while acquiring skin conductance responses (SCRs) ratings (arousal, valence, contingency). was operationalized as (1) dichotomization, following (2) specificity model, (3) cumulative risk (4) dimensional model. Individuals exposed to childhood showed blunted physiological reactivity SCRs, but not ratings, reduced CS+/CS- discrimination during both phases, mainly driven attenuated CS+ responding. The latter evident across different operationalizations ‘exposure’ theories. None tested clear explanatory superiority. Notably, remarkably pattern increased responding CS- reported literature for anxiety patients, suggesting that individuals may specific sub-sample. We highlight linking (vulnerability to) psychopathology need refinement.
