Abstract Having a hypoxic microenvironment is common and salient feature of most solid tumors. Hypoxia has profound effect on the biological behavior malignant phenotype cancer cells, mediates effects chemotherapy, radiotherapy, immunotherapy through complex mechanisms, closely associated with poor prognosis in various patients. Accumulating studies have demonstrated that normalization tumor vasculature, nanoparticle carriers biocarriers can effectively increase oxygen concentration microenvironment, improve drug delivery efficacy radiotherapy. They also infiltration innate adaptive anti-tumor immune cells to enhance immunotherapy. Furthermore, drugs targeting key genes hypoxia, including hypoxia tracers, hypoxia-activated prodrugs, hypoxia-inducible factors downstream targets, be used for visualization quantitative analysis antitumor activity. Developing a high-performance noninvasive probe for precise cancer theranostics is very challenging but urgently required. Herein, novel Au nanoclusters (NCs)-based was designed via ligand engineering by conjugating photoluminescent (PL) Au44 NCs in the second near-infrared window (NIR-II, 1000-1700 nm) with aromatic photoacoustic (PA)/photothermal molecules through click chemistry. This design bypasses incompatibility dilemma between photoluminescence attributes and PA/photothermal properties because rigidity of can lead to aggregation-induced emission (AIE) Au(i)-ligand shell constraining their nonradiative relaxation. Benefiting from strong NIR-II PL emissions at 1080 1240 nm, high photothermal conversion efficiency (65.12%), low cytotoxicity, appropriate renal clearance, enhanced permeability retention (EPR) effect, as-designed NC-based theranostic achieves ultradeep PL/PA imaging-guided therapy (PTT). Remarkably, 16 days after treatment guided imaging, mice were all healed without tumor recurrence, while average life span control groups only 17-21 days. study interesting it provides paradigm designing metal probe, may add fundamentally methodologically disease therapy.

Abstract The immunological implications of cuproptosis, a form cell death highly sensitive to oxygen presence, remain largely unexplored in the context tumor immunotherapy. Herein, we initially investigate positive correlation between cuproptosis and immunotherapy through bioinformatics analysis. Subsequently, an generator loaded with copper ions ( Cu/AP H‐M) has been constructed, which serves as effective carrier crucially enhances oxygenation microenvironment. The effectiveness of various cancer therapies for solid tumors is substantially limited by the highly hypoxic tumor microenvironment (TME). Here, a microalgae-integrated living hydrogel (ACG gel) developed to concurrently enhance hypoxia-constrained starvation therapy and immunotherapy. ACG gel formed in situ following intratumoral injection biohybrid fluid composed alginate, Chlorella sorokiniana, glucose oxidase, facilitated crossing-linking between divalent ions within alginate. microalgae sorokiniana embedded generate abundant oxygen through photosynthesis, enhancing oxidase-catalyzed consumption shifting TME from immunosuppressive immunopermissive status, thus reducing cell energy supply boosting antitumor immunity. In murine 4T1 models, significantly suppresses growth effectively prevents postoperative recurrence. This study, leveraging as natural oxygenerators, provides versatile universal strategy development oxygen-dependent therapies.
