Abstract Herein, we describe the design and development of a new cell‐permeable aggregation‐induced emission (AIE) active 3 ‐ethoxysalicylaldimine‐based symmetrical azine molecule HDBE . The synthesized compound underwent comprehensive investigation different spectroscopic methods, like NMR, mass single crystal X‐ray diffraction analysis. fluorophore exhibited bright orange colour AIE behaviour in THF–H 2 O mixture. drastic enhancement was achieved upon adding water to THF solution , with concentration 90%. Along dynamic light scattering (DLS) quantum yield measurements, formation aggregates also verified by scanning electron microscopy (SEM) transmission (TEM) Further, demonstrated excited state intramolecular proton transfer (ESIPT) characteristics polarity solvents, which corroborated absorption, lifetime spectroscopical investigations. detailed scrutiny structure displayed two strong hydrogen bonding interactions, while solid‐state fluorescent spectra showed dual that corresponds enol keto form confirming ESIPT feature. non‐toxic cell‐permeable, making it easy label as biomarker live HeLa cells via bioimaging. Acylaminoacyl-peptide hydrolase (APEH), a serine peptidase that belongs to the prolyl oligopeptidase (POP) family, catalyzes removal of N-terminal acetylated amino acid residues from peptides. As key regulator protein acetylation, APEH was involved in many important physiological processes while its aberrant expression correlated with progression various diseases such as inflammation, diabetics, Alzheimer's disease (AD), and cancers. However, emerging attention has been attracted APEH-related diagnosis drug discovery, mechanisms behind related are still unclear; thus, further investigating role function is great importance. To date, enzyme-activated fluorescent probes targeting POPs have extensively reported adopted relevant medical research applications. Nevertheless, an member POP rarely referred field bioimaging probe for vivo sensing activity not yet. Thus, acquiring efficient APEH-targeted urgent need. Herein, fluorogenic profiling first discovered via substrate mimic-based strategy. By combination stimulated molecular docking-based preliminary screening experiment-based secondary screening, optimal (named TMN-AcA), which displayed high binding affinity, sensitivity, specificity toward APEH, screened out.
