Carbazole/cyanobenzene photocatalysts promote the direct isotopic carboxylate exchange of C(sp3) acids with labeled CO2. Substrates that are not compatible transition-metal-catalyzed degradation–reconstruction approaches or prone to thermally induced reversible decarboxylation undergo incorporation at room temperature in short reaction times. The radiolabeling drug molecules and precursors [11C]CO2 is demonstrated.

18F labeling strategies for unmodified peptides with [18F]fluoride require 18F-labeled prosthetics bioconjugation more often cysteine thiols or lysine amines. Here we explore selective radical chemistry to target aromatic residues applying C–H 18F-trifluoromethylation. We report a one-step route [18F]CF3SO2NH4 from and its application direct [18F]CF3 incorporation at tryptophan tyrosine using as complex recombinant human insulin. The fully automated radiosynthesis of octreotide[Trp(2-CF218F)] enables in vivo positron emission tomography imaging.

While the development of positron emission tomography (PET) radiopharmaceuticals closely follows that traditional drug development, there are several key considerations in chemical and radiochemical synthesis, preclinical assessment, clinical translation PET radiotracers. As such, we outline fundamentals radiotracer design, with respect to selection an appropriate pharmacophore. These concepts will be reinforced by exemplary cases both their evaluation. We also provide a guideline for proper radionuclide labeling strategy access tracer optimal imaging qualities. Finally, summarize methodology evaluation vitro animal models road translation.
