Diabetes mellitus (DM) is a metabolic disorder that one of the most common health problems in world, primarily type 2 DM (T2DM). Metformin (MTF), as first-line treatment DMT2, effective lowering glucose levels, but its oral administration causes problems, including gastrointestinal side effects, low bioavailability, and risk hypoglycemia. In this study, we formulated MTF into microparticles incorporating glucose-responsive polymer (MP–MTF–GR), which could potentially increase bioavailability extend control release according to levels. This system was delivered by dissolving microneedles (MP–MTF–GR–DMN), applied through skin, thereby preventing effects orally administered MTF. MP–MTF–GR using various concentrations gelatin combined with phenylboronic acid (PBA) material. encapsulated DMN polyvinyl pyrrolidone (PVP) alcohol (PVA) polymers. The physicochemical evaluation showed be completely entrapped MP percentage trapped increasing concentration without changing chemical structure producing stable MP. addition, results MP–MTF–GR–DMN had adequate mechanical strength properties penetration ability environmental changes. vitro ex vivo permeation study on media from formula increased levels media. successfully skin at 11.30 ± 0.29, 23.31 1.64, 36.12 3.77, 53.09 3.01 μg PBS, PBS + 1%, 2%, 4%, respectively, 24 h, indicates behavior. developed also proven nontoxic based hemolysis tests. Importantly, rat model combination approach provide better reduction compared other routes, reducing blood level normal after 3 h maintaining for 8 h. Furthermore, did not change moisture rats. promising alternative delivery overcome effectiveness T2DM therapy.
