Targeted therapies for prostate, breast, and ovarian cancers are based on their activity against primary tumors rather than anti-metastatic activity. Consequently, there is an urgent need new agents targeting the metastatic process. Emerging evidence correlates in vitro vivo cancer invasion metastasis with increased of proteases mesotrypsin (prostate breast cancer) kallikrein 6 (KLK6; cancer). Thus, KLK6 attractive putative targets therapeutic intervention. As potential therapeutics advanced cancers, we report novel mesotrypsin- KLK6-based therapies, our previously developed mutants human amyloid β-protein precursor Kunitz protease inhibitor domain (APPI). These mutants, designated APPI-3M APPI-4M (ovarian cancer), demonstrated significant accumulation efficacy orthotopic preclinical models, advantages long retention times vivo, high affinity favorable pharmacokinetic properties. The applicability APPIs, as a therapy imaging purposes, supported by good safety profile controlled scalable manufacturability bioreactors.

Prostate cancer (PCa) recurrence affects between 20% and 40% of patients, being a significant challenge for predicting clinical outcomes increasing survival rates. Although serum PSA levels, Gleason score, tumor staging are sensitive detecting recurrence, they present low specificity. This study compared the performance three supervised machine learning models, Naive Bayes (NB), Support Vector Machine (SVM), Artificial Neural Network (ANN) classifying PCa events using dataset 489 patients from The Cancer Genome Atlas (TCGA). Besides comparing models performance, we searched analyzing whether incorporation specific genes expression in predictor set would enhance prediction then suggesting these as potential biomarkers patient prognosis. showed accuracy above 60% sensitivity 65% all combinations. ANN were more consistent their across different sets. Notably, SVM strong results precision specificity, particularly considering inclusion selected by feature selection ( NETO2 , AR HPN KLK3 ), without compromising sensitivity. However, relatively high standard deviations observed some metrics indicate variability simulations, gap additional studies via datasets.
