Human papillomaviruses (HPV) from the genus beta have been implicated in development of cutaneous squamous cell cancer epidermodysplasia verruciformis and organ transplant patients. In contrast to alpha-high-risk HPV, which cause ano-genital oropharyngeal cancers, beta-HPV replication is not well understood. The beta-HPV49 transcriptome was analyzed by RNA sequencing using stable keratinocyte lines maintaining high levels extrachromosomally replicating E8- genomes, can be established due a lack viral E8^E2 repressor protein. This analysis indicated presence four transcription start sites, two polyadenylation signals, splice donor (SD) acceptor sites consistent with conserved gene expression patterns animal human PV. Surprisingly, novel SD E6 oncogene (SD217) identified resembling carcinogenic alpha-HPV. Mutation SD217 enhanced protein but had no influence on growth keratinocytes transduced retroviral HPV49 E7 vectors. Inactivation context wild-type genome did enable immortalization prevented genome. mutant genomes revealed strong down-regulation usage, only weak effects other transcripts. suggests that does contribute modulating expression. Usage increased immortalized compared transiently transfected cells, may indicate long-term extrachromosomal maintenance requires reduced levels.IMPORTANCEHigh-risk (hr) alpha whereas skin hr-HPV, cycle very Transcriptional profiling reveals among HPV. site (SD217), previously described for controls oncoprotein required

Abstract PPP1R13L is a conserved inhibitor of p53, selectively regulating subset p53 target genes. Previous studies have reported that promotes cervical cancer progression, yet its precise mechanism remains unclear and warrants further investigation. In this study, we utilized public databases to reveal the correlation between tumor progression pathways. Subsequently, performed functional assays both in vitro xenograft models assess impact on cancer. Our results demonstrate cell proliferation, epithelial-mesenchymal transition, cycle glycolysis via PTEN/AKT/mTOR pathway. Mechanistically, regulates transcription PTEN through Ank-SH3 domain interaction with family, p63. 293T cells, originally exhibits significantly higher transcriptional activity than However, cancer—where E6 continuously degrades p63 highly expressed—p63 demonstrates for comparable to, or even surpasses, depending expression levels. Additionally, C33A, an HPV-negative line, R273C mutation causes exert opposite effect, shown be inhibited by independently p53. Finally, response elements PPP1R13L-regulated family genes were experimentally validated first time. This provides sequence basis selective regulation PPP1R13L. summary, underscore specificity PPP1R13L/p63/PTEN axis propose holds potential as therapeutic treatment. Graphical abstract

An examination of the fluctuating rates human papillomavirus (HPV) prevalence and allocation various genotypes could assist in implementation targeted strategies for prevention treatment cervical cancer. The present study was aimed at evaluating HPV genotype distribution among female populations Zhangjiagang, Jiangsu Province, China. A total 62,036 patients from Affiliated Zhangjiagang Hospital Soochow University provided clinical specimens between January 2022 August 2023, 12,144 women were infected with HPV, an overall positive rate 19.58%. most prevalent HR‐HPV types HPV52 (4.14%), HPV16 (2.44%), HPV53 (2.37%), HPV58 HPV56 (1.50%). In low‐risk group, highest infection HPV81 (1.76%). showed a “U”‐shaped curve. addition to adolescent other age groups are mainly single infection. And gynecology clinic group had much higher positivity compared physical ( p < 0.001). Our suggests that screening vaccination young highly necessary, 9‐valent vaccine is recommended regular immunization city.

Abstract Background There is a need for additional longitudinal studies with the Aptima messenger RNA human papillomavirus test (AHPV) to support safety of extended screening intervals. RNA‐based interventional nucleic acid (REINA) provides relevant information on clinical performance AHPV. Methods This prospective analysis 1538 participants after AHPV and liquid‐based cytology (LBC) co‐test complemented REINA protocol second 4 years negative 2000 women. Diagnostic accuracy cumulative risks CIN2+ up 9 were calculated all combinations. Results Sensitivity specificity 96.9% 88.0% 72.3% 92.0% LBC. Negative predictive value (NPV) positive (PPV) 99.9% 23.6%. The 5‐ 9‐year AHPV‐negative women 0.4% 1.0% (CIN2+) 0.3% 0.7% (CIN3+), 73% 64% lower risk than LBC ( p ≤ .002). an AHPV‐positive result at in first round had significantly 5‐year (11.1%) unknown HPV history (29.5%). Conclusions Currently, this constitutes longest European study testing population. It reveals NPV significant protective effect previous 5 new infection. These findings intervals beyond years. disease 3 High PPV may benefit controlling overtreatment colposcopy referrals.

MicroRNAs (miRNAs) are small non-coding RNAs (ncRNAs) that negatively regulate gene expression. MiRNAs fundamental biological processes and have significant roles in several pathologies, including cancer. Cervical cancer is the best-known example of a widespread human malignancy with demonstrated viral etiology. Infection high-risk papillomavirus (hrHPV) has been shown to be causative factor for cervical carcinogenesis. Despite occurrence prophylactic vaccines, highly sensitive HPV diagnostics, innovative new therapies, remains main cause death developing countries. The relationship between hrHPV infection depends on integration DNA host genome, disrupting regulator E2 continuous production E6 E7 proteins, which necessary acquire maintain transformed phenotype but insufficient malignant Lately, miRNAs, tumor microenvironment, immune evasion found major players carcinogenesis after infection. Many miRNAs widely reported as deregulated Here, relevance miRNA HPV-mediated transformation critically reviewed context natural history
