Polygonatum cyrtonema Hua is a perennial herbaceous plant of the genus, belonging to Liliaceae family, with significant medicinal and nutritional value. In China, this species traditional edible herb long history application widely appreciated by people. However, as demand for herbs continues grow, excessive harvesting has led depletion wild resources risk genetic erosion. addition, chaotic cultivation varieties lack high quality germplasm have inconsistent medical materials. Therefore, it urgent conduct diversity evaluation establish sound conservation plan. This study assessed population structure 96 samples collected from seven regions in China using simple sequence repeat (SSR) molecular marker technology. study, total 60 alleles (Na) were detected across 10 polymorphic SSR markers used, an average 6.0 generated per locus. The values information content (PIC) ranged 0.3396 0.8794, value 0.6430. effective number (Ne) was 2.761, Shannon's index (I) 1.196. analysis indicates that can be classified into three subpopulations (JZ, QY, JD) at level, which corresponds previous subgroups identified based on individual phenotypic traits. Analysis Molecular Variance (AMOVA) showed 74% variation between individuals within populations different regions. phylogenetic divided them main populations. QY JD are largely clustered together, could attributed their mountainous distribution local climate environment. differentiation coefficient (Fst) low 0.065, indicating relatively differentiation. ratio JZ other two (QY much higher than Based clustering results (Fst), inferred relationship closer, certain degree subpopulation. supports provides new parental material improvement breeding programs.

Cyanotoxins constitute a group of toxic secondary metabolites, the presence which in any water body poses major health risk. Moreover, advanced organisms such as edible plants exposed to these toxins, are possible pathway for human exposure. Green analytical chemistry is demanding environmentally friendly techniques. In this sense, we propose use capillary electrophoresis coupled tandem mass spectrometry (CE-MS/MS) determine mixture eight cyanotoxins belonging three different classes: cyclic peptides (microcystin-LR, microcystin-RR and nodularin), alkaloids (cylindrospermopsin anatoxin-a) isomeric non-protein amino acids (β-methylamino-L-alanine, 2,4-diaminobutyric acid N-(2-aminoethyl)glycine). Separation was achieved by using an acidic background electrolyte consisting 2 M formic 20 % acetonitrile water. Parameters affecting MS/MS detection sheath-liquid interface were also studied. Finally, combination pH-junction, field-amplified sample stacking (FASS) barrage online preconcentration strategies, employed improve sensitivity efficiency. The applied, with dual cartridge solid-phase extraction (SPE) system, allows obtain limits very low range µg·L−1 multiclass reservoir samples (from 0.005 0.10 µg·L−1). Furthermore, first time analysed spinach through CE-MS/MS same SPE procedure, following lyophilisation solid-liquid 6 mL 80% aqueous MeOH.
