Sirtuins are a family of proteins with enzymatic activity. There seven mammalian sirtuins (SIRT1-SIRT7) that found in different cellular compartments. They part crucial pathways and regulated by many factors, such as chemicals, environmental stress, phytochemicals. Several vitro vivo studies have presented their involvement anti-inflammatory, antioxidant, antiapoptotic processes. Recent findings imply phytochemicals resveratrol, curcumin, quercetin, fisetin, berberine, kaempferol may regulate the activity sirtuins. Resveratrol mainly activates SIRT1 indirectly AMPK. Curcumin influences SIRT3, but its is broad, cells affected. Quercetin modulates SIRT1, which triggers antioxidant responses. Fisetin, through regulation, modifies lipid metabolism anti-inflammatory Berberine has wide spectrum effects significant impact on signaling pathways. Finally, induction. This review aims to summarize recent properties modulation sirtuin activity, particular focus biochemical aspects.

Sirtuins (SIRT1-SIRT7), a family of nicotinamide adenine dinucleotide (NAD+)-dependent enzymes, are key regulators life span and metabolism. In addition to acting as deacetylates, some sirtuins have the properties deacylase, decrotonylase, adenosine diphosphate (ADP)-ribosyltransferase, lipoamidase, desuccinylase, demalonylase, deglutarylase, demyristolyase. Celiac disease (CeD), an autoimmune disorder triggered by gluten, affects around 1% of the global population. Standard treatment is a strict gluten-free diet (GFD), which poses significant challenges due to dietary restrictions, cross-contamination and subsequent persistent intestinal inflammation. This underscores need for new options addressing complex pathophysiology CeD. Recent research focuses on developing drugs that target barrier regeneration, gluten peptide modification, immune response alteration, gut microbial ecosystem modulation. These approaches offer potential more effective management CeD beyond GFD. Gluten-independent treatments may be particularly relevant under FDA's draft guidance CeD, emphasizes drug development as adjunct GFD patients with ongoing signs symptoms despite

ABSTRACT Background Cachexia is a wasting syndrome associated with imbalanced energy metabolism and loss of adipose muscle tissues contributes to morbidity mortality in ageing as well patients severe chronic diseases, including cancer. At present, there are no treatments addressing cachexia that have reached validation be used the clinic. In this study, we investigate protective role SIRT6, an important regulator homeostasis health preservation, against Lewis lung carcinoma (LLC)–induced cachexia. Methods SIRT6 levels serum from gastric cancer ( n = 22, 65.27 ± 12.50 years old, 40.9% females) healthy controls 63.50 10.77 45.4% were measured evaluate correlation between circulating development. Ten‐week‐old transgenic (TG) wild type (WT) male mice injected LLC cells (1.5 × 10 6 per mouse) effects on cachexia‐associated browning lipolysis underlying mechanisms. We explored effect LLC‐conditioned medium induced mature adipocytes, differentiated primary mouse embryonic fibroblasts (MEFs). evaluated vitro activator by treatment MDL800. Results concentrations significantly higher non‐cachectic (3.41 0.30 ng/mL) compared cachectic (3.20 0.23 ng/mL, p < 0.01), suggesting negative level overexpression ameliorated tumour‐induced expenditure white (eWAT mass loss: 66% WT vs. 32% TG; iWAT 69% 40% TG) through suppression lipolysis. LLC‐induced cachexia, tumour necrosis factor‐α receptor 2 (TNFR2) mediated inhibition lipolytic signalling, because difference knockout group was almost eliminated TNFR2 neutralizing antibody. Increased concentration found (690.41 pg/mL 1166.98 patients, 0.05). A selective pharmaceutical activator, MDL800, could completely reverse adipocytes. SIRT6 belongs to the conserved NAD+-dependent deacetylase superfamily and mediates multiple biological pathological processes. Targeting by allosteric modulators represents a novel direction for therapeutics, which can overcome selectivity problem caused structural similarity of orthosteric sites among deacetylases. Here, developing reversed strategy AlloReverse, we identified cryptic site, Pocket Z, was only induced bi-directional signal triggered upon binding NAD+. Based on discovered an inhibitor named JYQ-42. JYQ-42 selectively targets other histone deacetylases effectively inhibits deacetylation, with IC50 2.33 μmol/L. significantly suppresses SIRT6-mediated cancer cell migration pro-inflammatory cytokine production. JYQ-42, our knowledge, is most potent selective inhibitor. This study provides drug design will help in challenging development therapeutic agents that bind SIRT6.
