Use of potent painkillers like opiates are limited by their abuse potential and adverse physiological effects necessitating new therapeutics for pain management. This study assessed the efficacy oral cannabinoid formulations (F1-F4) in alleviating chronic neuropathic (CP) investigated mechanisms through thermal algesia, inflammatory oxidative stress biomarkers, sensory nerve conduction velocity (SNCV). A 21-day rat model constriction injury (CCI) sciatic was used to evaluate (F1: 500 mg, F2: 1000 F3: 2000 F4: 3000 mg) MCT oil, with pregabalin as reference. Male Wistar rats (35) were divided equally into seven groups, all except Sham group undergoing ligation receiving different formulations.On day 22, behavioral (hot plate, tail flick) electrophysiological (sensory velocity, SNCV) assessments performed. SNCV also measured presence CB1 CB2 receptor antagonists. Additionally, blood-based markers inflammation (TNF-α) (MDA, GSH CAT) analysed.
