Abstract Platelet‐derived growth factors (PDGFs) and receptors (PDGFR) play a key role in the process of coronary atherosclerosis. We aimed to investigate association genetic variations mRNA expressions PDGF/PDGFRB pathway genes with artery disease (CAD). In this case–control study (3139 CAD vs. 3270 controls), 13 single nucleotide polymorphisms (SNPs) at five were genotyped combined construct weighted risk score (wGRS). Three hundred six pairs cases controls selected for quantification. Restricted cubic spline (RCS) analyses conducted dose–response relationship between wGRS, mRNAs CAD. BackgroundChuanzhi Qingyu (CZQY) recipe is a traditional Chinese medicine clinically used for the secondary prevention and treatment of cardiovascular diseases associated with atherosclerosis (AS) However, potential active components underlying molecular mechanisms require further elucidation.PurposeThis study aims to explore CZQY their action in stabilizing vulnerable AS plaques.MethodsInitially, mouse model plaques was established by inducing conditions high-fat diet. The efficacy these assessed using histological staining techniques, including HE, Oil Red O, Masson staining. Additionally, biochemical analyses, ELISA assays, small animal ultrasound technology, immunohistochemistry were employed. Subsequently, pharmaceutical ingredients identified through UPLC-Q-TOF-MSE comparing them standard reference substances. Finally, network pharmacology approach, conjunction RNA-seq utilized predict targets plaques, which subsequently validated Western blotting quantitative PCR methods.ResultsThe has been demonstrated significantly improve lipid liver function, reduce serum levels inflammation oxidative stress, markedly lower critical biomarkers plaques. Pathological indicate that decreases plaque area, accumulation while increasing collagen fiber ratio α-SMA CD68. Ultrasound assessments reveal improves vascular narrowing elasticity. Network analyses revealed primary stabilize include flavonoids, monoterpenes, diterpenes, phenolic compounds. These are involved regulation metabolism, inflammatory responses, stress extracellular matrix dynamics, cytokine activity, platelet function. analysis aorta produced results consistent findings from pharmacology. qPCR modulated protein mRNA key targets.ConclusionCZQY effectively reduces risk factors stabilizes This may be related its compounds such as terpenoids, organic acids, modulation inflammation, cell adhesion atherosclerosis. provide significant data support pharmacological research clinical applications.

Atherosclerotic cardiovascular disease poses a significant health challenge globally. Recent findings highlight the pivotal role of endothelial-to-mesenchymal transition (EndMT) in atherosclerosis. Morin is bioflavonoid mainly extracted from white mulberry, traditional Chinese herbal medicine with anti-inflammatory and antioxidant properties. This study examines whether morin can alleviate atherosclerosis by suppressing EndMT seeks to elucidate underlying mechanism.
