The onset of Alzheimer's disease (AD) is significantly influenced by oxidative stress. Trace element selenium (Se) can be incorporated into selenoproteins to participate in redox regulation. In our previous study, it was found that brain-targeting chlorogenic acid-modified nanoparticles (SeNPs) could effectively relieve stress AD. However, the main mechanism polyphenol-decorated SeNPs on antioxidant enzymes such as glutathione peroxidase still unknown. Dihydromyricetin (DMY) one polyphenol has neuroprotective effects through activities. this chitosan (CS) introduced a stabilizer for synthesis brain-targeted peptide (Tg: TGNYKALHPHNG) and DMY-modified (Tg-CS/DMY@SeNPs). Tg-CS/DMY@SeNPs improve solubility DMY exhibited better free radical scavenging ability than DMY. Meanwhile, enhanced activity enzymes, which inhibited ROS accumulation Aβ aggregate-induced PC12 cells. More importantly, activate Nrf2/Keap-1 signaling pathway regulate especially selenophenase brain liver APP/PS1 mice. case, also prevented damage-induced apoptosis inhibiting Caspase-3 Finally, remarkably improved cognitive disorders These results indicated promising therapeutic therapy

Abstract The abnormal accumulation of β ‐amyloid protein (A ) is considered as the main pathological hallmark Alzheimer's disease (AD). design potent multifunctional theranostic agents targeting A one effective strategies for AD prevention and treatment. Nanomotors intelligent, advanced, nanoplatforms can perform many complex tasks, but their application in theranostics rare. Herein, sub‐10nm dual‐carbon dots composites (ERCD) with photo‐propelled nanomotor behavior are fabricated by conjugating near‐infrared (NIR) carbon (RCD) thermogenic photodynamic capability previously reported epigallocatechin gallate‐derived carbonized polymer (ECD). ERCD‐1 (ECD:RCD = 1:2.5) showed inhibitory similar to ECD absence NIR light, exhibited photooxygenation activity powered “self‐thermophoretic force” under irradiation, significantly enhancing inhibition, disaggregation, capabilities. suppressed fibrillization rapidly disaggregated mature fibrils at very low concentrations (0.5 µg mL −1 ). Moreover, NIR‐activated imaged plaques vivo prolonged nematode lifespan 6 d 2 . As a proof‐of‐concept, this work opened new avenue nanomotors theranostics.
