Idiopathic pulmonary fibrosis (IPF) is a fatal lung disease, characterized by distorted architecture and loss of respiratory function. Alveolar epithelial cell injury hyperplasia, enhanced extracellular matrix deposition, (myo)fibroblast activation are features IPF. Wnt/beta-catenin signaling has been shown to determine fate during development. As aberrant reactivation developmental pathways suggested contribute IPF pathogenesis, we hypothesized that activated in cells Thus, quantified localized the expression activity pathway IPF.The Wnt1, 3a, 7b, 10b, Wnt receptors Fzd1-4, Lrp5-6, as well intracellular signal transducers Gsk-3beta, beta-catenin, Tcf1, 3, 4, Lef1 was analyzed transplant donor lungs quantitative real-time (q)RT-PCR. 7b Fzd2 significantly increased Immunohistochemical analysis Wnt3a, Gsk-3beta largely alveolar bronchial epithelium. This confirmed qRT-PCR primary type II (ATII) cells, demonstrating significant increase ATII derived from patients. In addition, Western blot phospho-Gsk-3beta, phospho-Lrp6, target genes cyclin D1, Mmp 7, or Fibronectin 1 demonstrated functional compared with controls. Functional vitro studies further revealed ligands induced proliferation collagen synthesis. Our study demonstrates expressed operative adult Increased may be involved impaired epithelial-mesenchymal cross-talk modification represent therapeutic option

Alzheimer disease (AD) is the most common form of age-related dementia. The etiology AD considered to be multifactorial as only a negligible percentage cases have familial or genetic origin. Glycogen synthase kinase-3 (GSK-3) regarded critical molecular link between two histopathological hallmarks disease, namely senile plaques and neurofibrillary tangles. In this review, we summarize current data regarding involvement kinase in several aspects development progression, well key observations highlighting GSK-3 one relevant targets for treatment.

Protein phosphorylation controls many aspects of cell fate and is often deregulated in pathological conditions. Several recent findings have provided an intriguing insight into the spatial regulation protein across different subcellular compartments how this can be finely orchestrated by specific kinases phosphatases. In review, focus will placed on (i) phosphoinositide 3-kinase (PI3K) pathway, specifically Akt mTOR phosphatases PP2a PTEN, (ii) PKC family serine/threonine kinases. We look at general physiology controlled these phosphatases, highlighting signalling pathways that drive division, proliferation, apoptosis.

Although glycogen synthase kinase-3 beta (GSK-3 β ) was originally named for its ability to phosphorylate and regulate glucose metabolism, this multifunctional kinase is presently known be a key regulator of wide range cellular functions. GSK-3 involved in modulating variety functions including cell signaling, growth various transcription factors that determine the survival or death organism. Secondary role diseases Alzheimer’s disease, inflammation, diabetes, cancer, small molecule inhibitors are gaining significant attention. This paper primarily focused on addressing bifunctional conflicting roles both promotion apoptosis. has emerged as an important molecular target drug development.
