ABSTRACT Bergapten (BP) is a plant‐derived furocoumarin that has wide range of pharmacological effects. BP serves as candidate amplifier in phototherapy against skin inflammation, such psoriasis and atopic dermatitis. However, the anti‐inflammatory role remains elusive. We utilized IL‐17A‐stimulated keratinocyte line imiquimod‐challenged BALB/c mice to imitate psoriasis‐like inflammation. Inflammatory phenotypes were determined by expressions inflammatory genes cytokines, histopathological changes activities nuclear factor‐κB (NF‐κB) pathway. An RNA‐seq analysis rodent was performed explore possible mechanism lying behind. SiRNAs antagonist (TMS) cytochrome P450 family 1 subfamily B member (CYP1B1) subsequently used determine CYP1B1 pathogenesis vitro vivo. Overexpression with lentivirus further validate therapeutic effect BP. significantly suppressed activation NF‐κB pathway inhibiting p65 phosphorylation improved phenotype both revealed key regulating signaling Knock‐down siRNAs reduce expression cytokines. intraperitoneal injection TMS partially remediated IMQ‐induced mainly terms thickness. Cyp1b1 led increased protein rescued vitro. This study keratinocytes inhibited blocking p65.
