Abstract Phospholipase D (PLD) is a valuable enzyme in industrial processes for converting phosphatidylcholine (PC) to phosphatidylserine (PS). In this study, strain of Bacillus cereus was isolated from soil and identified through 16S rRNA sequencing. To enhance PLD activity, various mutagenesis strategies—including chemical treatment, irradiation, their combinations—were employed, resulting four high-activity positive mutants (C-7, I-12, CI 7–12, IC 13–14). Among these, the 7–12 mutant exhibited significantly higher enzymatic showing 3.12-fold increase (312.2%) compared wild-type strain. Fermentation conditions were optimied using response surface methodology (RSM), achieving activity 35 U/mL. The demonstrated stability over wide temperature range (30–60°C) pH (6–10), with half-life 128 days. Kinetic analysis revealed Vmax 20.04 µmol/h Km 7.13 µmol/mL, indicating efficient activity. bioconversion experiments, PLD-enriched fermentation broth catalyzed conversion PC PS, 53.0% rate 92.3% selectivity PS two-phase system. Agarwood, a precious traditional medicinal herb and fragrant material, is known for its sedative sleep-improving properties. This study explores the mechanisms underlying aromatherapy effects of Chi-Nan agarwood ordinary in improving sleep. Using combination gas chromatography–mass spectrometry (GC-MS), network pharmacology, molecular docking techniques, we identified c ompared chemical compositions potential targets both types agarwood.
