Gastrointestinal diseases are global health issues. Current drugs for gastrointestinal cause discomfort and toxicity; consequently, the use of traditional medicines their extracts has gained attention in recent years. Physochlaina physaloides (L) G. Don. (P. physaloides) is traditionally used diarrhoea gastroenteritis; however, its material basis mechanism action gastric injury have not been fully studied. This study aims to explore P. protective effects on injury, together with potential mechanisms. We constructed chronic gastritis ulcer models rats using 56 % ethanol anhydrous ethanol, respectively. Additionally, we screened pathways via transcriptomics gene expression omnibus (GEO) database. Subsequently, an ethanol-stimulated GES-1 cell model active fraction based survival rate antioxidant activity. The effect was investigated tissue structure (HE staining), mucus secretion (PAS anti-inflammatory activity, acid levels. employed liquid chromatography-tandem mass spectrometry (LC-MS/MS) determine components drug blood, before investigating mechanisms immunofluorescence, transcriptomic, metabolomics, network pharmacology, molecular docking, qRT-PCR, western blotting, flow cytometry. occurrence gastritis, ulcer, cancer related PPAR/NF-κB signalling pathway, decreased FABP3 PPARγ, increased Bcl-2 TNF-α. n-butanol (BPP) showed significant improvement activity vitro. BPP also alleviated inflammation oxidative stress rat models, including by upregulating CAT, GSH, SOD, IL-10, PGE2; downregulating VEGFA, IL-6, IL-8, TNF-α, NO; improving pathological damage; restoring levels; reducing macrophage expression. components, anisodamine hyoscyamine, upregulated PPARα, CPT1, FABP3, downregulated NF-κB p65, thereby regulating pathway gastroprotection. did significantly increase GPX4 SLC7A11, nor they reduce production ROS. Therefore, unrelated ferroptosis. provides first evidence effectiveness prevention ulcers treatment gastritis. adopted a multidisciplinary approach demonstrate that protect against ethanol-induced non-ferroptotic death pathways. can target PPARγ may clinical application prospects prevent which unique advantages. These findings provide scientific foundation gastroprotection expand applications BPP.
