Abstract Background Hepatocellular carcinoma (HCC) is one of the most common malignant tumors with a poor prognosis. Cuproptosis recently confirmed novel kind programmed cell death and associated variety cancer. However, prognostic value cuproptosis-related genes (CRGs) still limited in HCC. Methods Gene expression data clinical information HCC patients were downloaded from Cancer Genome Altas (TCGA) Gene-Expression Omnibus (GEO) databases. Differential analysis was performed to identify differentially expressed CRGs. Then univariate multivariate Cox regression analyses applied screen out prognosis-related CRGs construct CRG models. The Kaplan-Meier survival Receiver operating characteristic (ROC) curves employed assess values model. Cibersort used evaluate relationship between model immune infiltration. In addition, Set Enrichment Analysis (GSEA) explore molecular mechanism Results A total 120 screened. containing three (ABCB6, AACS, CKAP2) constructed for independently predicting prognosis showed that high-risk group had significantly shorter overall (OS). ROC indicated better than single gene rates. Univariate demonstrated an independent predictor. Moreover, correlated stromal score, tumor purity T cells folicular helper, regulator, macrophages M0. GSEA revealed cycle, oocyte meiosis, mitotic nuclear division, DNA replication pathways mainly enriched group. Conclusion This study HCC, which has high predictive efficacy. These findings extend knowledge provide theoretical support prediction, may inform new therapeutic strategies

Abstract Long non-coding RNAs (lncRNAs) have emerged as crucial regulators in various cellular processes, including cancer progression and stress response. Recent studies demonstrated that copper accumulation induces a unique form of cell death known cuproptosis, with lncRNAs playing key role regulating cuproptosis-associated pathways. These may trigger cell-specific responses to stress, presenting new opportunities prognostic markers therapeutic targets. This paper delves into the cuproptosis-mediated cancer, underscoring their potential biomarkers targets for innovative strategies. A thorough review scientific literature was conducted, utilizing databases such PubMed, Google Scholar, ScienceDirect, search terms like 'lncRNAs,' 'cuproptosis,' 'cancer.' Studies were selected based on relevance lncRNA regulation cuproptosis pathways implications prognosis treatment. The highlights significant contribution cuproptosis-related genes pathways, impacting metabolism, mitochondrial responses, apoptotic signaling. Specific are breast, lung, liver, ovarian, pancreatic, gastric cancers. objective this article is explore cancers mediated by cuproptosis.

Ubiquitination-related genes (URGs) exerted a crucial part in variety of human disease disorders; however, their association with pancreatic adenocarcinoma (PAAD) had yet to be clearly described. We aimed comprehensively characterize the contributions URGs PAAD through silico analysis and experimental validation, then identified robust mRNA-lncRNA-based molecular prognostic panel for patients using bulk RNA-sequencing single-cell data. Initially, we collected multi-omics data from TCGA platform depict comprehensive landscape pan-cancer. Furthermore, were accurate in-depth analysis. Significant differences activation ubiquitination pathways expression detected between normal malignant cells. Unsupervised hierarchical clustering determined two subtypes distinct clinical outcomes, pathway activities, immune microenvironment, functional annotation characteristics. The profiles ubiquitination-associated mRNAs lncRNAs training validation datasets utilized develop verify novel ubiquitination-related mRNA-lncRNA panel, which satisfied prediction efficiency. Our model could function as an effective index outperformed four other recognized panels evaluating patients' survival status. Tumor mutation burden, chemotherapy response intensively explored demonstrate underlying mechanism difference according our panel. findings also revealed that FTI-277, farnesyltransferase inhibitor, better curative effect high-risk patients, while MK-2206, Akt allosteric superior therapeutic low-risk patients. real-time PCR results uncovered RNA AC005062.1 all three cell lines was elevated several thousandfold. In conclusion, URGs-based classification triumphantly served tool evaluation PAAD, this developed potential target therapy.

Iron copper are essential metal ions critical maintaining cellular function. The accumulation of iron triggers distinct death pathways, known as ferroptosis cuproptosis, respectively. Ferroptosis characterized by iron-dependent lipid peroxidation, while cuproptosis involves copper-induced oxidative stress. They increasingly recognized promising targets development anticancer drugs. Recently, compelling evidence demonstrated that interplay between plays a crucial role progression. This review elucidates converging pathways Moreover, we examined value genes associated with clinical diagnosis treatment cancer, mainly outlining potential co-targeting approach.
