Nasopharyngeal carcinoma (NPC) presents significant treatment challenges due to its complex etiology and late-stage diagnosis. The traditional Chinese medicine Selaginella doederleinii Hieron (S. doederleinii) has shown potentiality in NPC multi-target, multi-pathway anti-cancer mechanisms. First, we identified related target genes from databases like GeneCards, OMIM, DisGeNET, performed WGCNA analysis on the GSE53819 dataset identify several important gene modules NPC. Active components their targets S. were screened TCMSP other databases, identifying 32 overlapping genes. Gene Ontology (GO) Kyoto Encyclopedia of Genes Genomes (KEGG) pathway revealed that these are primarily involved critical biological processes protein phosphorylation cell cycle regulation. A protein–protein interaction network was constructed, cytoHubba six key (BCL2, MAPK14, ABCB1, PLK1, ATM, HMOX1). Kaplan–Meier immune infiltration further showed closely prognosis microenvironment patients. Abstract Nasopharyngeal carcinoma (NPC) is an Asia‐prevalent malignancy, yet its genetic underpinnings remain incompletely understood. Here, a transcriptome‐wide association study (TWAS) conducted on NPC, leveraging gene expression prediction models based epithelial tissues and genome‐wide (GWAS) summary statistics from 1577 NPC cases 6359 controls of southern Chinese descent. The TWAS identifies VAMP8 chromosome 2p11.2 as novel susceptibility for NPC. Further fine‐mapping analyses pinpoint rs1058588, located within , causal variant through eQTL colocalization, GWAS across multiple cohorts, achieving significance (OR = 1.18, P 3.09 × 10 −10 ). Functional assays demonstrate that exerts tumorigenic role in enhancing cell proliferation, migration, tumor growth. Mechanically, it uncovered rs1058588 modulates by altering binding affinity to miR‐185 . Furthermore, the results show interacts with DHX9 facilitate nuclear recruitment p65, activating NF‐κB pathway. Collectively, findings shed light predisposition underscore critical functional axis involving miR‐185, VAMP8, DHX9, pathway pathogenesis.

Squalene epoxidase (SQLE), a key enzyme in cholesterol metabolism, remains underexplored nasopharyngeal carcinoma (NPC). Additionally, the therapeutic potential of Fangchinoline, an alkaloid with anticancer properties, has yet to be systematically evaluated. Immune checkpoint inhibitors (ICIs), particularly PD-1/PD-L1 inhibitors, have demonstrated significant survival benefits in treating recurrent or metastatic nasopharyngeal carcinoma (R/M-NPC). While baseline peripheral blood lymphocyte subsets been identified as prognostic biomarkers various cancers treated with ICIs, their relevance R/M-NPC has not extensively studied. This post hoc analysis used data from 153 patients PD-L1 inhibitor monotherapy the phase 2 trial KL167-2-05-CTP. The subsets, including total T cells, CD4/CD8 ratio, helper suppressor cytotoxic NK and B were tested by flow cytometry.
