The conservation of the genetic resources old trees is crucial to their ecological role but extremely difficult, especially for oak species (Quercus spp.) displaying recalcitrance in seed and vegetative propagation methods. Our study aimed assess regenerative potential Quercus robur different ages (up 800 years) during micropropagation. We also determine how vitro conditions can influence regeneration responses. Lignified branches collected from 67 selected were cultivated ex culture pots at 25 °C obtain epicormic shoots (explant sources). explants on an agar medium supplemented with 0.8 mg L-1 6-benzylaminopurine (BAP) least 21 months. Bacopa floribunda (Family: Plantaginaceae) is used in folklore medicines for the management of cognitive dysfunction. It has nootropic, antiaging, anti-inflammatory, anti-cholinesterase, and antioxidant properties. We developed an efficient reproducible protocol vitro propagation B. using nodal explants. assessed effects Murashige Skoog (MS) medium fortified with various plant growth regulatory substances (PGRs), a precursor, elicitors their optimal combinations on regeneration production total saponins, triterpenoid saponin glycosides (bacoside A3, bacopaside X, II, bacosaponin C), stigmasterol content grown biomass floribunda. Abstract Background Quercus seeds that are recalcitrant to desiccation and freezing temperatures cannot be stored in gene banks under conventional conditions. However, the germplasm of some seeded species can liquid nitrogen (–196 °C). Unfortunately, for many species, among them almost whole genus , an effective cryostorage method is still unknown. In this study, we propose a successful protocol petraea (Matt.) Liebl. using plumules (a shoot apical meristem embryo) frozen on aluminium cryo-plates. Results The isolated from acorns ten provenances were prestored 0.5 M sucrose solution (for 18 h). To form alginate beads (one plumule per bead), placed wells cryo-plate embedded calcium gel. Abstract In the in vitro cloning of specific individuals woody species, a challenge can be low viability explants collected from old mature donor trees due to recalcitrance their plant material. Other factors, such as auxin–cytokinin balance, must also considered. We investigated how conditions and age modulate regeneration capacity, morphophysiology, DNA methylation Quercus robur . Explants two different donors (70- 600-year-old trees) were subjected 8 treatments, which consisted 0, 1.25, 3.5, or 7.0 µM 6-benzylaminopurine (BAP) combined with concentrations 0 0.5 1-naphthaleneacetic acid (NAA). absence BAP, there was no shoot induction, root formation achieved only shoots 70-year-old clone. Shoot number lower Both clones showed increased capacity under BAP exposure, positively correlated 8-hydroxydeoxyguanosine (8-oxo-dG) levels antioxidant total phenolics. contrast, induction negatively 5-methylcytosine (m 5 C) levels. Treatments induced development anatomical biochemical characteristics underdeveloped tissues and/or delayed tissue development, this effect less intense Global hypomethylation strongly correlates guanine hyperoxidation de novo response. Our results suggest that fully may have growth regulator supplementation needs ‘donor age’ factor. Older require higher concentration cytokinins ensure regenerative
