The conservation of the genetic resources old trees is crucial to their ecological role but extremely difficult, especially for oak species (Quercus spp.) displaying recalcitrance in seed and vegetative propagation methods. Our study aimed assess regenerative potential Quercus robur different ages (up 800 years) during micropropagation. We also determine how vitro conditions can influence regeneration responses. Bacopa floribunda (Family: Plantaginaceae) is used in folklore medicines for the management of cognitive dysfunction. It has nootropic, antiaging, anti-inflammatory, anti-cholinesterase, and antioxidant properties. Abstract Background Quercus seeds that are recalcitrant to desiccation and freezing temperatures cannot be stored in gene banks under conventional conditions. However, the germplasm of some seeded species can liquid nitrogen (–196 °C). Unfortunately, for many species, among them almost whole genus , an effective cryostorage method is still unknown. In this study, we propose a successful protocol petraea (Matt.) Liebl. using plumules (a shoot apical meristem embryo) frozen on aluminium cryo-plates. Results The isolated from acorns ten provenances were prestored 0.5 M sucrose solution (for 18 h). To form alginate beads (one plumule per bead), placed wells cryo-plate embedded calcium gel. For cryoprotection, encapsulated immersed cryoprotectant containing 2.0 glycerol different concentrations (0.8–1.2 M) 40 min at 25 °C desiccated laminar flow cabinet 1.0–4.0 h. Cryo-plates with directly then cryostored 30 min. rewarming, cryo-plates 1.0 rehydrated 15 °C. Survival rates varied 25.8 83.4 achieved after cryoprotection drying 2 vitro regrowth rate cryopreserved was 26–77%. Conclusions This study presents, first time, successful, simple cryopreservation Q. could used banks. experiment successfully repeated various provenances, each yielding similar, good results. seed quality storage time harvesting important factors cryopreservation.
