Gastric cancer remains one of the most prevalent and lethal malignancies worldwide, characterized by poor survival rates, particularly in advanced stages. In recent years, a paradigm shift gastric treatment has been witnessed with introduction immunotherapy targeted therapies. This review provides detailed examination current immunotherapeutic strategies, including adoptive cell therapy (ACT), immune checkpoint inhibitors (ICIs), vaccines. Additionally, it explores advancements therapies, focusing on human epidermal growth factor receptor 2 (HER2) vascular endothelial (VEGFR) signaling pathways, as well emerging targets such claudin 18.2. Clinical trials investigating chimeric antigen T-cell (CAR-T) therapy, receptor-engineered (TCR-T) natural killer (NK) cell-based treatments have shown promise, when combined conventional chemotherapeutic regimens. However, challenges cytokine release syndrome, immune-related toxicities, scalability issues remain significant. The combination therapies represents promising approach to enhance outcomes. Future directions emphasize need overcome resistance mechanisms refine strategies improve efficacy while reducing adverse effects. aims elucidate landscape explore their potential shaping future clinical management for this devastating disease.

The exploration of cancer vaccines has yielded a multitude studies, resulting in diverse collection information. heterogeneity vaccine data significantly impedes effective integration and analysis. While CanVaxKB serves as pioneering database for over 670 manually annotated vaccines, it is important to distinguish that database, on its own, does not offer the structured relationships standardized definitions found an ontology. Recognizing this, we expanded Vaccine Ontology (VO) include those present were initially covered, enhancing VO's capacity systematically define interrelate vaccines. An ontology design pattern (ODP) was first developed applied semantically represent various capturing their associated entities relations. By applying ODP, generated template tabular format converted into RDF/OWL generation terms VO. '12MP vaccine' used example demonstrate application ODP. VO also reuses reference such diseases hosts. Therapeutic vaccines containing aluminum adjuvants have been widely used in the treatment of tumors due to their powerful immune-enhancing effects. However, neurotoxicity with different physicochemical properties has not completely elucidated. In this study, a library engineered oxyhydroxide (EAO) and hydroxyphosphate (EAHP) nanoparticles was synthesized determine vitro. It demonstrated that surface charge EAHPs size EAOs did affect cytotoxicity N9, bEnd.3, HT22 cells; however, soluble ions trigger three cell lines. Moreover, induce apoptosis N9 cells, further mechanistic studies mediated by mitochondrial reactive oxygen species generation membrane potential loss. This study identifies safety profile aluminum-containing salts nervous system during therapeutic vaccine use, provides novel design strategies for safer applications. SIGNIFICANCE STATEMENT: it cells. comparation, triggered significant lines, indicating form which is presenting may play crucial role its safety. induced dependent on damage. confirms formulations.
