Sessile plants engage in trade-offs between growth and defense capacity response to fluctuating environmental cues. MYB is an important transcription factor that plays many roles controlling plant defense. However, the mechanism behind how it keeps a balance these two physiological processes still largely unknown. Our work focuses on dissection of molecular by which GhMYB33 regulates The CRISPR/Cas9 technique was used generate mutants for deciphering functions. Sphingolipids serve as essential components of biomembrane and possess significant bioactive properties. Sphingosine-1-phophate (S1P) plays a key role in plant resistance to stress, but its specific impact on growth development remains be fully elucidated. Cotton fiber cells are an ideal material for investigating the maturation cells. In this study, we examined content composition sphingosine (Sph) S1P throughout progression cell development. The elevated gradually during elongation declined transition stage. Exogenous application promoted while using FTY720 (an antagonist S1P), DMS inhibitor LCBK) hindered elongation. Long Chain Base Kinase 1 (GhLCBK1) was notably expressed stage, containing all conserved domains LCBK protein localized endoplasmic reticulum. Overexpression GhLCBK1 increased retarded secondary wall (SCW) deposition. Conversely, downregulation reduced levels, suppressed elongation, accelerated SCW Transcriptome analysis revealed that upregulating or applying induced expression GhEXPANSIN auxin related genes. Furthermore, levels IAA were fibers when up-regulating down-regulating GhLCBK1, respectively. Our investigation demonstrated product facilitated by affecting biosynthesis. This study contributes novel insights into intricate regulatory pathways involved identifying potential target gene laying groundwork enhancing quality via genetic manipulation.
