Modulation of immune microenvironment is critical for inflammatory bowel disease (IBD) intervention. Epigallocatechin gallate (EGCG), as a natural low toxicity product, has shown promise in treating IBD. However, whether and how EGCG regulates the intestinal not fully understood. Here we report that lessens colitis by orchestrating Th1 polarization self-amplification novel manner required multilevel-regulated microecosystem. Mechanistically, activates GPR43 on IEC to inhibit dependently short chain fatty acid (SCFA)-producing gut microbiota. Inhibition activity weakens protective effects development. Moreover, confirm fecal SCFAs and/or are limited patients with correlated cell number. Taken together, our study reveals an microenvironment-dependent immunoregulatory IBD provides insight into mechanisms EGCG-based immunotherapeutic strategies

Carvacrol and thymol (CAT) have been widely recognized for their antimicrobial anti-inflammatory properties, yet specific effects on colitis the mechanisms involved remain insufficiently understood. This study establishes a causative link between CAT administration mitigation, primarily through enhancement of Bifidobacterium pseudolongum abundance in colon. increase promotes production secondary bile acids, particularly hyodeoxycholic acid (HDCA) 12-ketodeoxycholic (12-KCAC), which exert effects. Notably, does not alleviate symptoms germ-free mice, indicating necessity gut microbiota. research uncovers novel regulatory mechanism where HDCA 12-KCAC inhibit colonic inflammation by reducing expression transmembrane guanylate cyclase 1A epithelium. downregulation elevates intracellular Ca
