Nanocarriers Targeting Circular RNA ADARB1 Boost Radiosensitivity of Nasopharyngeal Carcinoma through Synergically Promoting Ferroptosis

Nasopharyngeal
carcinoma
(NPC)
is
a
common
malignant
tumor
of
the
head
and
neck,
prevalent
in
regions
such
as
Southern
China
Southeast
Asia.
Radiotherapy
serves
primary
clinical
treatment
for
this
carcinoma.
However,
resistance
to
radiotherapy
fundamental
cause
failure
patient
mortality,
with
underlying
mechanisms
yet
be
fully
elucidated.
We
identified
recently
characterized
circular
RNA,
circADARB1,
which
markedly
upregulated
NPC
tissues
closely
associated
poor
prognosis
resistance.
Both
vitro
vivo
experiments
demonstrated
that
circADARB1
inhibited
ferroptosis,
thereby
inducing
cells.
Building
on
these
findings,
we
synthesized
biomimetic
nanomaterial
consisting
semiconducting
polymer
nanoparticles
wrapped
cell
membranes,
designed
deliver
both
siRNA
targeting
iron
ions.
The
application
not
only
efficiently
suppressed
expression
boosted
intracellular
concentrations,
but
also
enhanced
ferroptosis
induced
by
radiotherapy,
improving
radiosensitivity
Furthermore,
our
study
revealed
heat
shock
protein
HSP90B1,
repaired
misfolded
SLC7A11
GPX4
proteins
triggered
preserving
their
stability
biological
functions.
Mechanistically,
facilitated
cysteine
transportation
into
cells
glutathione
synthesis,
while
employed
mitigate
lipid
peroxidation
shielding
from
oxidative
damage
inhibiting
ultimately
leading

investigation
elucidates
molecular
substantial
relevance,
highlights
promising
prospects
nanotechnology
precision
cancer
therapy.
