Abstract Diaporthe is an important plant pathogenic genus, which also occurs as endophytes and saprobes. Many species that are morphologically similar proved to be genetically distinct. The current understanding of taxonomy by applying morphological characters, host associations multi-gene phylogeny problematic leading overestimation/underestimation numbers this significant fungal genus. Currently, there no definite boundaries for the accepted species. Hence, present study aims re-structure genus , based on single gene phylogenies (ITS, tef tub cal his ), justified GCPSR (Genealogical Concordance Phylogenetic Species Recognition) methodology well coalescence-based models (PTP—Poisson Tree Processes mPTP—multi-rate Poisson Processes). Considering all available type isolates divided into seven sections while 13 15 species-complexes proposed. To support re-assessment 82 obtained from woody hosts in Guizhou Province China were investigated revealed presence two novel 17 previously known Synonymies specified 31 molecular data studies. Dividing several specific phylogenetic analyses can avoid construction lengthy trees entire future taxonomic In other words, when one conducts research related only appropriate section need selected analysis.

Considering that fungi display a great morphological, ecological, metabolic, and phylogenetic diversity, their taxonomic identification is extremely important because it helps us establish information about each species its possible biochemical ecological roles. Traditionally, the of at level has been carried out with molecular tools such as DNA sequencing, but still represents huge challenge today due to heterogeneity fungal kingdom, making task complex difficult process. Biotyping, type chemotaxonomy, developed in field identification/differentiation classification micro-fungi through mass spectrometry (MS). Here, two endophytic strains isolated from different hosts were cultivated studied regarding morphology biology. Morphology analysis determined Diaporthe, results grouped them D. melongenae. We sought faster less way differentiating these interest an MS chemical profile MS/MS data using low-resolution spectrometer. Additionally, we linked this structure compounds previously genus Diaporthe. Studies conducted technique allowed propose distinctive molecules are unique strain share common (13 total). In addition, first report secondary metabolites The dataset demonstrates under investigation can be distinguished via spectrometry, suggesting host affinity; both exhibits pronounced differences profiles across all culture media incubation periods parameters described herein.
