ABSTRACT Heart failure with preserved ejection fraction (HFpEF) accounts for approximately 50% of heart cases globally, and this incidence is increasing due to extended lifespans accumulating comorbidities. Emerging evidence suggests that Wnt signaling plays a role in cardiomyocyte hypertrophy cardiac fibrosis, which are key features HFpEF. Furthermore, Porcupine (PORCN) inhibitors, negatively regulate signaling, have shown promising results improving function reducing and/or fibrosis. This study investigated whether acute oral administration the PORCN inhibitor, Wnt‐c59, alters maladaptive structural functional mouse model Male mice were given high‐fat diet L‐NAME (0.5 g L −1 ) drinking water 5 weeks, followed by 2‐week intervention orally administered Wnt‐c59 (5 mg kg day ). HFpEF characterized hypertension, diastolic dysfunction, although there was no activation heart. Despite this, short‐term treatment ameliorated adverse remodeling ratio more compliant collagen type 3 tensile 1 also improved associated increased expression brain natriuretic peptide, known promoter ventricular compliance.
