Arsenic has been ranked as the most hazardous substance by U.S. Agency for Toxic Substances and Disease Registry. Environmental arsenic exposure-evoked health risks have become a vital public concern worldwide owing to widespread existence of arsenic. Multi-omics is revolutionary technique data analysis providing an integrated view bioinformation comprehensively systematically understanding elaborate mechanism diseases. This study aimed at uncovering potential contribution liver-microbiota-gut axis in chronic inorganic exposure-triggered biotoxicity chickens based on multi-omics technologies. Forty Hy-Line W-80 laying hens were chronically exposed sodium arsenite with dose-dependent manner (administered drinking water containing 10, 20, or 30 mg/L arsenic, respectively) 42 d, followed transcriptomics, serum non-targeted metabolome, 16S ribosomal RNA gene sequencing accordingly. intervention induced serious chicken liver dysfunction, especially severe fibrosis, simultaneously altered ileal microbiota populations, impaired intestinal barrier, further drove enterogenous lipopolysaccharides translocation via portal vein circulation aggravating damage. Furtherly, injured disturbed bile acids (BAs) homoeostasis through strongly up-regulating BAs synthesis key rate-limiting enzyme CYP7A1, inducing excessive total accumulation, accompanied massive primary BA—chenodeoxycholic acid. Moreover, concentrations secondary BAs—ursodeoxycholic acid lithocholic markedly repressed, which might involve repressed dehydroxylation Ruminococcaceae Lachnospiraceae families. Abnormal metabolism turn promoted injury, ultimately perpetuating pernicious circle chickens. Notably, obvious depletion abundance four profitable microbiota, Christensenellaceae, Ruminococcaceae, Muribaculaceae, Faecalibacterium, correlated tightly this hepato-intestinal process Our demonstrates that exposure evokes disruption establishes scientific basis evaluating risk environmental pollutant

Deltamethrin (DLM) is a widely used and highly effective insecticide. DLM exposure harmful to animal human. Quail, as bird model, has been in the field of toxicology. However, there little information available literature about quail cerebrum damage caused by DLM. Here, we investigated effect on neurons. Four groups healthy quails were assigned (10 each group), respectively given 0, 15, 30, 45 mg/kg gavage for 12 weeks. Through measurements cerebrum, it was found that induced obvious histological changes, oxidative stress, neurons apoptosis. To further explore possible molecular mechanisms, performed real-time quantitative PCR detect expression endoplasmic reticulum (ER) stress-related mRNA such glucose regulated protein 78 kD, activating transcription factor 6, inositol requiring enzyme, kinase RNA (PKR)-like ER kinase. In addition, detected ATP content evaluate functional status mitochondria. The study showed significantly increased decreased tissues. These results suggest chronic induces apoptosis via promoting stress mitochondrial dysfunction. Furthermore, our provide novel explanation DLM-induced avian

Poria cocos (P. cocos) has been traditionally used as folk medicine and functional food in China for more than 2000 years. The water-soluble polysaccharide is the main component of P. decoction. effects mechanisms from (PCWP) were investigated chronic sleep deprivation (CSD)-induced anxiety rats. CSD induced anxiety, gut dysbiosis, inflammatory responses, reduced neurotransmitter levels, whereas PCWP intervention ameliorated anxiety-like behaviors, increased levels 5-hydroxytryptamine, dopamine, norepinephrine, γ-aminobutyric acid hypothalamus, regulated gastrointestinal peptide factors, inhibited tumor necrosis factor (TNF)-α/nuclear (NF)-κB signaling pathway rats with CSD. changes intestinal flora composition determined using 16S rDNA sequencing, indicated that significantly improved species richness diversity adjusted abundance following dysregulated bacteria closer to normal group: Rikenellaceae_RC9_gut_group, Ruminococcus, Prevotellaceae_UCG-001, Prevotellaceae_NK3B31_group, Fusicatenibacter. Metabolomics was analyze fecal samples identify altered metabolites PCWP-treated groups. Thirty-eight PCWP-related four metabolic pathways such sphingolipid metabolism, taurine hypotaurine vitamin B6 glycerophospholipid metabolism explored. results serum metabolomics showed 26 biomarkers changed after compared model group. regulatory enrichment on sphingolipid, phenylalanine, validation activity enzymes involved above pathways. A strong correlation between potential found. Our findings present new evidence supporting effect preventing progression by inhibiting TNF-α/NF-κB pathway, alleviating disorders, ameliorating microflora imbalance.

The indiscriminate use of deltamethrin in agriculture and aquaculture can lead to residues increased many regions, which poses negative impacts on intestinal health aquatic organisms. Although the potential toxicity have recently attracted attention, comprehensive studies injuries after chronic exposure remain poorly understood. Herein, a 28-day test, crucian carp expose different concentrations (0, 0.3, 0.6 μg/L) were used as research object. We found that morphology changes goblet cells tissue, extent tissue injury along with increasing dose deltamethrin. Additionally, genes expression antioxidant activity (Cu/Zn superoxide dismutase (Cu-Zn SOD), glutathione peroxidase 1 (GPX1), catalase (CAT)), inflammatory response (tumor necrosis factor alpha (TNFα), interferon gamma (IFNγ), interleukin beta (IL-1β)), tight junctions (Claudin 12 (CLDN12), junction protein (ZO-1)) dramatically increased. Meanwhile, apoptosis autophagy process triggered through caspase-9 cascade related 5 (ATG5)- (ATG12) conjugate. Besides, amount Proteobacteria Verrucomicrobiota, while decreased Fusobacteriota abundance, resulting microbiota function disorders. In summary, our results highlight cause serious physiological flora disturbances.
