Antibiotic resistance is currently one of the most significant threats to global public health and safety. And studies have found that over next 25 years, 39 million people will die directly 169 indirectly due antibiotic-resistant diseases. Consequently, development new types antimicrobial drugs urgently needed. Antimicrobial peptides (AMPs) constitute an essential component innate immune response in all organisms. They exhibit a distinctive mechanism action endows them with broad spectrum biological activities, including antimicrobial, antibiofilm, antiviral, anti-inflammatory effects. However, AMPs also present certain limitations, such as cytotoxicity, susceptibility protein hydrolysis, poor pharmacokinetic properties, which impeded their clinical application. The delivery systems can address these challenges by modifying AMP enabling precise, controlled release at site infection or disease. This review offers comprehensive analysis mechanisms advantages AMPs. systematically evaluate how emerging drug systems, nanoparticles hydrogels, enhance stability bioavailability AMPs, discussing both strengths limitations. Moreover, unlike previous reviews, this highlight recent clinically approved AMP-based those development, emphasizing key translating into practice. With perspectives, it hoped provide some insights overcoming translational barriers advancing

The prevalence of multidrug resistance (MDR) and stagnant drug-development pipelines have led to the rapid rise hard-to-treat antibiotic-resistant bacterial infections. These infectious diseases are no longer just nosocomial but also becoming community-acquired. spread MDR has reached a crisis level that needs immediate attention. landmark O’Neill report projects by 2050, mortality rates associated with infections will surpass individuals afflicted cancer. Since conventional antimicrobials very reliable, it is great importance investigate different strategies combat these life-threatening diseases. Here, we provide an overview recent advances in viable alternative treatment mainly targeting pathogen’s virulence capability rather than viability. Topics include small molecule immune inhibition factors, quorum sensing (QS) quenching, biofilm development, bacteriophage-mediated therapy, manipulation individual’s macroflora

Periodontitis is a chronic inflammatory condition driven by plaque-associated microorganisms, where uncontrolled bacterial invasion and proliferation impair host immune responses, leading to localized periodontal tissue inflammation bone destruction. Conventional therapies face challenges, including incomplete microbial clearance the rise of antibiotic resistance, limiting their precision effectiveness in managing periodontitis. Recently, nanotherapies based on polymeric materials have introduced advanced approaches antimicrobial therapy through diverse mechanisms. ABSTRACT Objectives The rise in carbapenem‐resistant Enterobacterales (CRE) has reinforced the global quest for developing effective therapeutics. Traditional drug discovery approaches have been inadequate overcoming this challenge due to their resource and time constraints. Methods English literature was searched by structured queries related our review between January 1, 2020, December 31, 2024. Results key resistance mechanisms CRE, such as enzymatic hydrolysis, decreased permeability, efflux pump overexpression, examined review. Abstract The compound sulfamethoxazole (SMZ)/trimethoprim (TMP), a dual‐agent formulation comprising SMZ and TMP, exhibits synergistic bacteriostatic bactericidal activity by disrupting folate metabolism pathways. This study assessed the bioequivalence (BE) of generic SMZ/TMP tablet versus its innovator counterpart under fasting conditions (n = 24). In meticulous, single‐site, randomized, open‐label, 2‐period, 2‐sequence crossover trial, 24 healthy Chinese adults were allocated to receive single administration either test or reference medication, with 7‐day interval between doses. Venous blood samples obtained pre‐dose at intervals up 48 hours postdose for subsequent analysis. plasma levels TMP determined using validated ultra‐performance liquid chromatography‐tandem mass spectrometry technique. Safety monitoring was conducted precision throughout trial all subjects. study's results indicated no significant differences in peak concentrations (C max ) drug when comparing 2 formulations. Furthermore, 90% confidence ratios geometric means C , area curve from 0 time last quantifiable concentration point (AUC 0‐t ), extrapolated an infinite 0‐∞ within BE range accepted as 80%‐125%.
