Background: Nibribacter radioresistens, a UV and gamma radiation-tolerant bacterium, was reported to have superior antibacterial activities against variety of pathogenic bacteria through the production antimicrobial peptides (AMPs), but nothing is known about its AMPs. Methods/Results: In this study, our genomic transcriptomic data showed that N. radioresistens genome contains 11 AMP gene candidates, designated as NB_AMP1 NB_AMP11, which are expressed differently in logarithmic growth stationary phase. Moreover, cell-free supernatant all Escherichia coli DH5α strains containing cloned AMPs except for NB_AMP5 NB_AMP7 exhibited both Gram-negative Gram-positive such E. Staphylococcus aureus. Synthetic supported AMPs, and, particular, synthetic NB_AMP2 S. Conclusions: Altogether, these results suggest candidates from may function peptides, effectively causing cellular lysis pore formation bacterial membrane.

Antimicrobial peptides (AMPs) come from various sources and exhibit unique antimicrobial properties. Their rapid action, effectiveness, resistance to development make them promising alternatives combat antibiotic resistance. In addition its excellent antibacterial properties, AMPs have superior immunomodulatory, antitumor, antiviral activities. recent years, the demand for has continued increase in many fields, especially medical field, prospects are extensive. However, disadvantages of expensive cost, higher hemolysis, short half-life, susceptibility degradation by protein hydrolases, low bioavailability, toxic side effects, other disadvantages, which seriously limit wide application AMPs. Therefore, fewer been approved marketing or undergoing clinical trials. The review covers period 2001 2025 provides a detailed discussion searching databases such as Google Scholar Web Science. This paper reviews progress research on sources, structures, optimization strategies, biological activities, mechanisms applications. general, approaches number new increased significantly. improvement technologies high poor stability, bioavailability cost is still high, but widely used clinical, food, livestock, poultry, cosmetics fields. article focuses commonly strategies main activities AMPs, aiming effectively respond challenges provide theoretical basis expanding their range.

Background/Objectives: Microbial infections represent a significant threat to public health due the emergence and spread of antimicrobial resistance. Adjunctive alternative therapeutic strategies are explored tackle this issue, including use natural or synthetic peptides. Previous research showed that antibody-derived peptides possess antimicrobial, antiviral, immunomodulatory properties. This study aimed characterize newly designed evaluate their effectiveness against representative strains Staphylococcus aureus, drug-resistant isolates. Methods: Colony-forming unit assays confocal microscopy studies were performed peptide activity planktonic microbial cells. Cytotoxicity tests on THP-1 human monocytic Circular dichroism (CD) nuclear magnetic resonance (NMR) employed for conformational characterization Results: The half-maximal effective concentrations bacterial reference isolates ranged from 0.17 18.05 µM, while cytotoxic effects not observed mammalian A killing kinetics analysis observation by interaction between bacteria suggested mechanism action involving membrane perturbation. CD all predominantly exhibit random coil arrangement in aqueous solution. NMR spectroscopy revealed most active adopts helical conformation presence mimetics. Conclusions: structural evaluation peptides’ may lead selection candidate be further studied develop an treatment caused strains.
