Abstract DNA nanostructures have shown great potential in biomedical fields. However, the immune responses, especially activation of cGAS‐STING signaling (A‐cGSs), induced by nanostructures, remain incompletely understood. Here, ability various from double‐stranded (dsDNA), single‐stranded tiles (SSTs) to origami is investigated on A‐cGSs. Unlike natural dsDNA which triggers potent A‐cGSs, structural interconnectivity configurations can substantially reduce occurrence irrespective their form, dimensions, and conformation. wireframe activate signaling, suggesting that decreasing A‐cGSs compactness‐dependent. Viruses have evolved multiple mechanisms to counteract the stimulator of interferon genes (STING) pathway, resulting in suppression antiviral responses. Accordingly, addition developing STING agonist analogs with enhanced stability and deliverability, overcoming defective function virus-infected cells is essential for defense against viral infections. In this study, we developed pathway-activating complexes (SPAC) based on bioinspired vesicles that display multimerize molecules a specific affinity binding. As broad-spectrum agent, universal mimic triggers IFN-I signaling independently endogenous STING. infectious models, including CMV SARS-CoV-2 infection, both prophylactic therapeutic regimens SPAC can reduce load disease severity. These results indicate SPAC, functioning as host-targeted immune modulator, provides distinct advantage therapy diseases caused by DNA RNA viruses, particularly those strong antagonistic functions. The STING-independent activation may provide diseases, potentially serving candidate option defend future pandemics "Disease X".
