Abstract Background: Periosteum plays an important role in bone defect repair due to its rich vascular network and cells. However, natural periosteum is difficult meet clinical requirements low availability. Therefore, it necessary develop a tissue engineering strategy of biomimetic for repair. Methods: Poly-ε-caprolactone/chitosan/Whitlockite electrospun bionic membrane (PCL/CS/WH) was prepared using electrospinning technology, then conjugated with E7 peptide as PCL/CS/WH/E7 membrane. The physical properties the membranes were evaluated by TEM FTIR. In vitro, LIVE/DEAD staining Cell Counting Kit-8 assay marrow mesenchymal stem cells (BMSCs) Endothelial progenitor (EPCs) are used assess biocompatibility membranes. Matrigel applied evaluate ability different composite nanofibers samples promote angiogenesis. Mineralized nodule collagen formation BMSCs detected alizarin red sirius respectively. vivo, regeneration angiogenesis assessed Micro-CT associated staining. Result: The addition chitosan (CS) (E7) enhanced hydrophilicity cytocompatibility pure PCL Additionally, CS, Mg2+ released from Whitlockite (WH) had synergistic effect osteogenic differentiation. Three weeks after implantation, successfully bridged significantly promoted new blood vessels. Concultion: achieve efficient enriches solutions treatment critical defects.

Osteoporosis is a systemic skeletal disease caused by an imbalance between bone resorption and formation. Current treatments primarily involve medication hormone therapy. However, these lack directionality are often ineffective for locally severe osteoporosis, with the potential complex adverse reactions. Consequently, treatment strategies using bioactive materials or external interventions have emerged as most promising approaches. This review proposes twelve microenvironmental targets osteoporosis-related pathological changes, including local accumulation of inflammatory factors reactive oxygen species (ROS), mitochondrial dynamics, insulin resistance, disruption cell autophagy, apoptosis, changes in neural secretions, aging cells, increased tissue vascular destruction, decreased regeneration. Additionally, this examines current research status effective biophysical biochemical stimuli based on summarizes advantages optimal parameters different bioengineering to support preclinical clinical osteoporosis Finally, addresses ongoing challenges future prospects.

Although silicon is a widespread constituent in dental materials, its possible influence on the formation and repair of teeth remains largely unexplored. Here, we studied effect two silicic acid-releasing nanomaterials, silica bioglass, living model pulp consisting stem cells seeded dense type I collagen hydrogels. Silica nanoparticles released acid had little cell viability mineralization efficiency but impacted metabolic activity, delayed matrix remodeling, led to heterogeneous distribution. Bioglass improved activity homogeneous dispersion mineral deposits within hydrogel. These results suggest that presence calcium ions bioglass not only favorable proliferation can also counterbalance negative effects silicon. Both chemical biological processes should therefore be considered when investigating silicon-containing materials tissues.

Reconstruction of extensive bone defects due to age, trauma, or post-illness conditions remains challenging. Biomimetic scaffolds with osteogenic capabilities have been proposed as an alternative the classical autograft and allograft implants. Three-dimensional were obtained based on Ce-doped mesoporous bioactive glass (MBG) Spongia agaricina (SA) sacrificial templates functionalized vitamin D3. The study aimed investigate effect D3 functionalization optimal variant a 3D scaffold doped 3 mol% ceria, selected in our previous work its biological physicochemical properties. Scanning electron microscopy (SEM) images non-functionalized/functionalized revealed porous structure interconnected pores ranging from 100 350 μm. Fourier transform infrared spectroscopy (FTIR) SEM analysis confirmed surface functionalization. Cytotoxicity evaluation showed that all investigated do not exhibit cytotoxicity genotoxicity toward Saos-2 osteosarcoma cell line. Moreover, demonstrated enhanced activity dental pulp stem cells (DPSCs) by increasing calcium deposition osteocalcin secretion, determined Alizarin red stain colorimetric ELISA kit, result synergistic action cerium ions. results MBG had potential for applications regeneration.
