Spermatozoa are highly specialized male cells that characterized by a unique ability to move, which is critical factor for successful fertilization. The relative simplicity of motility assessment, especially in livestock, has made it widely used parameter evaluating ejaculate quality or cryopreserved semen the clinical field, and an advanced tool reproductive physiology toxicology research. Technological advances image analysis computational methods have substantially increased its accuracy through use computer-assisted sperm (CASA) minimize subjective bias assessments. Nevertheless, this more objective method still presents some significant challenges, including variability sample preparation, imaging conditions, analytical parameters. These issues contribute inconsistency impair reproducibility comparability data between laboratories. implementation standardized protocols, combined with comprehensive training rigorous evaluation, can serve mitigate emerging inconsistencies. In addition, vitro conditions under CASA analyses performed often differ significantly from natural environment female tract vivo. This review discusses methodologies, issues, limitations using CASA, particular focus on boar as important agricultural biomedical model species system used.

ABSTRACT Semen analysis is an important laboratory diagnostic test for stallions. Evaluation of sperm motility integral to basic semen and results are breeding management clinical practice. Computer‐assisted (CASA) allows objective evaluation increases analytical precision. The the present study was validate a portable CASA system (AndroScope). Fresh/cooled samples (33 ejaculates, 18 stallions) frozen (40 ejaculates one epididymal flush, 27 were evaluated using AndroScope IVOS II (‘gold standard’). This study aimed to characterize the sperm kinematic values with high frames per second, define subpopulation structure of a horse and donkey compare them. A total 57 fresh semen ejaculates (26 Spanish 16 Arabian breeds 10 donkeys) were collected subsequently analyzed for parameters using Computer-aided motility analysis ISAS®v1.2 system Spermtrack® 10-μm depth counting chamber. Sequences recorded at 250 eight automatically evaluated. All showed significant differences between breeds. evaluated excellent characteristics, significantly higher all donkeys compared horses except beat-cross frequency. Donkey was faster linear than horse. Regarding differences, had average path velocity, curvilinear frequency rapid, but Arab more linear. The principal component three subpopulations in ejaculate different characteristic dominant both straight, beat (38.2 41.7%, respectively), whereas lowest slowest non-linear sperms. This, plus slight distribution these horses, found. In conclusion, permitted have new interpretation motile species breed differences.
