Vitamin D is a natural photoproduct that has many beneficial effects on different organs, including skin. Active forms of vitamin and its derivatives exert biological skin cells, thus maintaining homeostasis. In keratinocytes, they inhibit proliferation stimulate differentiation, have anti-inflammatory properties, act as antioxidants, DNA damage repair after ultraviolet (UV) exposure. melanocytes, also cell proliferation, apoptosis antioxidants. fibroblasts, affect fibrotic processes collagen production, promote wound healing regeneration. On the other hand, cells ability to activate directly. These activities, along with projected topical application derivatives, are promising for care photo protection can be used in prevention or possible reversal aging.

ABSTRACT The aryl hydrocarbon receptor (AhR), a ligand‐activated transcription factor, is extensively expressed in diverse human organs and plays pivotal role mediating the onset, progression, severity of numerous diseases. Recent research has explored substantial impact AhR on skin homeostasis related pathologies. As multi‐layered organ, comprises multiple cell populations that express AhR. In this review, we introduce various cells its barrier function. Furthermore, explore involvement development diseases, highlighting potential as therapeutic target for disorders. By targeting AhR, may open new avenues novel efficient disease treatments.

Background/Objectives: Voglibose, an α-glucosidase inhibitor commonly prescribed to manage postprandial hyperglycemia in diabetes mellitus, demonstrates potential for repurposing as anti-melanogenic agent. This study aims explore the inhibitory effects of voglibose on melanogenesis and elucidate its molecular mechanisms, highlighting possible applications treating hyperpigmentation disorders. Methods: The were investigated using B16F10 melanoma cells. Cell viability, melanin content, tyrosinase activity assessed following treatment. Western blot analysis was performed examine changes melanogenic proteins transcription factors. role signaling pathways, including PKA/CREB, MAPK, PI3K/AKT, GSK3β/β-Catenin, analyzed. Primary human skin irritation tests conducted evaluate topical safety voglibose. Results: Voglibose significantly reduced synthesis cells a dose-dependent manner. revealed decreased expression MITF, TRP-1, TRP-2, indicating inhibition melanogenesis. modulated key suppression AKT activation, while restoring GSK3β inhibit β-catenin stabilization. Human confirmed voglibose’s application, showing no adverse reactions at 50 100 μM concentrations. Conclusions: properties through modulation multiple pathways biosynthesis. Its profile efficacy suggest repurposed drug managing advancing cosmeceutical applications.

The skin–brain axis has been suggested to play a role in several pathophysiological conditions, including opioid addiction, Parkinson’s disease and many others. Recent evidence suggests that pathways regulating skin pigmentation may directly indirectly regulate behaviour. Conversely, CNS-driven neural hormonal responses have demonstrated pigmentation, e.g., under stress. Additionally, due the shared neuroectodermal origins of melanocytes neurons CNS, certain CNS diseases be linked pigmentation-related changes common regulators, MC1R variations. Furthermore, HPA analogue connects endocrine system, thereby allowing index possible abnormalities visibly. In this review, insight is provided into pigment production neuromelanin synthesis brain recent findings are summarised on how signalling skin, with particular focus interconnected central nervous system. Thus, review supply better understanding mechanism associations health disease.
