Depression is a common and serious mental disorder. Data on its pathogenesis remain unclear the options of drug treatments are limited. Here, we explored role pyroptosis, novel pro-inflammatory programmed cell death process, in depression as well anti-depression effects mechanisms salidroside (Sal), bioactive extract from Rhodiola rosea L. We established corticosterone (CORT)-induced or lipopolysaccharide (LPS)-induced mice vivo, CORT, nigericin (NLRP3 agonist)-induced PC12 cells vitro. Our findings demonstrated that Sal profoundly mediated CORT LPS-induced depressive behavior improved synaptic plasticity by upregulating expression brain-derived neurotrophic factor (BDNF) gene. The data showed upregulation proteins associated with NLRP3-mediated including NLRP3, cleaved Caspase-1, IL-1β, IL-18, GSDMD. molecular docking simulation predicted would interact P2X7 P2X7/NF-κB/NLRP3 signaling pathway. In addition, our pyroptosis was regulated Interestingly, shown to ameliorate via suppression rescued nigericin-induced cells. Besides, knock down NLRP3 gene siRNA markedly increased inhibitory proinflammatory responses. Taken together, plays crucial depression, ameliorates suppressing P2X7/NF-κB/NLRP3-mediated pyroptosis. Thus, study provides new insights into potential treatment for depression.
