Methods: In this study, Puerarin -loaded nanoparticles (Pue-NPs) were prepared using the cell-penetrating peptide-poly (lactic-co-glycolic acid) (CPP-PLGA) as drug carrier by emulsified solvent evaporation method. Then, they added into poly (acrylic to obtain self-assembled nanocomposite hydrogels (SANHs) delivery system co-polymerization The particle size, zeta potential, micromorphology Pue-NPs measured; appearance, mechanical properties, adhesive strength, biological activity SANHs performed. Metabolic syndrome (MetS) is a cluster of risk factors for cardiovascular diseases (CVDs) that has become global public health problem. Puerarin (PUE), the principal active compound Pueraria lobata, effects regulating glucose and lipid metabolism protecting against damage. This study aimed to investigate whether dietary supplementation with PUE could ameliorate MetS its associated Rats were randomly divided into three groups: normal diet group (NC), high-fat/high-sucrose (HFHS), HFHS plus (HFHS-PUE). The results showed PUE-supplemented rats exhibited enhanced tolerance, improved parameters, reduced blood pressure compared those on alone. Additionally, reversed HFHS-induced elevations in atherogenic index (AI) activities serum lactate dehydrogenase (LDH) creatine kinase (CK). Ultrasonic evaluations indicated significantly ameliorated cardiac dysfunction arterial stiffness. Histopathological assessments further confirmed mitigated remodeling, neuronal damage brain. Moreover, lowered systemic inflammatory indices including C-reactive protein (CRP), neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte (MLR), immune-inflammation (SII).
