The P2X7 receptor (P2X7R), a non-selective cation channel modulated by adenosine triphosphate (ATP), localizes to microglia, astrocytes, oligodendrocytes, and neurons in the central nervous system, with most incredible abundance microglia. P2X7R partake various signaling pathways, engaging immune response, release of neurotransmitters, oxidative stress, cell division, programmed death. When neurodegenerative diseases result neuronal apoptosis necrosis, ATP activates P2X7R. This activation induces biologically active molecules such as pro-inflammatory cytokines, chemokines, proteases, reactive oxygen species, excitotoxic glutamate/ATP. Subsequently, this leads neuroinflammation, which exacerbates involvement. is essential development diseases. implies that it has potential drug target could be treated using antagonists are able cross blood-brain barrier. review will comprehensively objectively discuss recent research breakthroughs on genes, their structural features, functional properties, roles possible therapies.

Background: Delayed wound healing in skin injuries has become a significant problem clinics, seriously affecting and even threatening life health. Recently, research interest increased developing dressings containing bioactive compounds capable of improving outcomes for complex needs. Methods: In this study, Puerarin -loaded nanoparticles (Pue-NPs) were prepared using the cell-penetrating peptide-poly (lactic-co-glycolic acid) (CPP-PLGA) as drug carrier by emulsified solvent evaporation method. Then, they added into poly (acrylic to obtain self-assembled nanocomposite hydrogels (SANHs) delivery system co-polymerization The particle size, zeta potential, micromorphology Pue-NPs measured; appearance, mechanical properties, adhesive strength, biological activity SANHs performed. Finally, potential was further evaluated streptozotocin-induced diabetic mice. Results: regularly spherical, with an average size 134.57 ± 1.42 nm 2.14 0.78 mV. colorless transparent honeycomb-like porous structure had excellent swelling ratio (917%), water vapor transmission rate (3077 g·m − 2 ·day 1 ), properties (Young's modulus 18 kPa, elongation at break 307%), strength (15.5 kPa). exhibited sustained release Pue over 48h, cumulative 55.60 6.01%. vitro tests revealed that presented 92.22% antibacterial against Escherichia coli after 4h, 61.91% scavenging 1.1-diphenyl-2-trinitrophenylhydrazine (DPPH) radical. vivo experiments showed accelerated repair reducing inflammatory response site, promoting angiogenesis, facilitating epidermal regeneration collagen deposition. Conclusion: conclusion, we successfully SANHs. Our results show have performance improves mice model, indicating it can be used develop effective strategy treatment wounds. Alzheimer's disease (AD) is the most common neurodegenerative disease, which mainly caused by damage of structure and function central nervous system. At present, there are many adverse reactions in market-available drugs, can't significantly inhibit occurrence AD. Therefore, current focus research to find safe effective therapeutic drugs improve clinical treatment Oxidative stress bridges different mechanism hypotheses AD plays a key role Numerous studies have shown that natural flavonoids good antioxidant effects. They can directly or indirectly resist oxidative stress, Aβ aggregation Tau protein hyperphosphorylation activating Nrf2 other oxidation-antioxidation-related signals, regulating synaptic function-related pathways, promoting mitochondrial autophagy, etc., play neuroprotective In this review, we summarised inhibiting oxidative injury recent years. Moreover, because shortcomings poor biofilm permeability low bioavailability flavonoids, advantages progress nano-drug delivery systems such as liposomes solid lipid nanoparticles were highlighted. We hope review provides useful way explore treatments.

Metabolic syndrome (MetS) is a cluster of risk factors for cardiovascular diseases (CVDs) that has become global public health problem. Puerarin (PUE), the principal active compound Pueraria lobata, effects regulating glucose and lipid metabolism protecting against damage. This study aimed to investigate whether dietary supplementation with PUE could ameliorate MetS its associated Rats were randomly divided into three groups: normal diet group (NC), high-fat/high-sucrose (HFHS), HFHS plus (HFHS-PUE). The results showed PUE-supplemented rats exhibited enhanced tolerance, improved parameters, reduced blood pressure compared those on alone. Additionally, reversed HFHS-induced elevations in atherogenic index (AI) activities serum lactate dehydrogenase (LDH) creatine kinase (CK). Ultrasonic evaluations indicated significantly ameliorated cardiac dysfunction arterial stiffness. Histopathological assessments further confirmed mitigated remodeling, neuronal damage brain. Moreover, lowered systemic inflammatory indices including C-reactive protein (CRP), neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte (MLR), immune-inflammation (SII). In conclusion, effectively moderated metabolic disorders, attenuated inflammation, minimized induced by an diet. These provide novel insights potential benefits prevention management related CVDs.

Abstract Hashimoto’s thyroiditis (HT) is the most common organ-specific autoimmune disease, predominantly affecting women. Although pathogenesis of HT incompletely understood, some studies have found that macrophage polarization plays a role. Puerarin soy isoflavone compound has anti-inflammatory and immunomodulatory effects regulates immune activity. This study aimed to verify therapeutic effect puerarin on explored its regulatory imbalance in HT. Through bioinformatics analysis molecular biology methods, it was macrophages increased significantly patients model mice. Immunological staining showed intervention could reduce tissue inflammatory cell infiltration. Molecular biological examination displayed inhibit local systemic inflammation levels, expression marker thyroglobulin thyroid peroxidase Abs. In vivo experimental results indicated regulated polarity reduced damage, possibly by inhibiting pyroptosis signaling pathway. clearance experiments demonstrated relied exert mechanism action treating The this indicate are important mediators development HT, can regulate status provide protection, which provides new idea for treatment
