Background Liver cancer's severity has prompted interest in herbal remedies for hepatotoxicity and the disease. Acacia nilotica (Vachellia nilotica), valued its traditional medicinal properties, particularly aerial components, is being investigated hepatoprotective anti-cancer effects. However, research on A. bark extracts their mechanisms of action limited. Materials Methods The study aimed to determine Total Phenolic Content (TPC), Flavonoid (TFC), antioxidant properties extract using biochemical assays. Additionally, it examined protective effects normal Cells (LO2) toxicity against liver cancer cells (HepG2) cell culture techniques. Results showed significant (EC50-16.34), with a total phenolic content 159.98±9.91 mg GAE/DW flavonoid 16.93 QE/g dry weight. It exhibited moderate towards HepG2 at concentrations >100 μg/mL but was non-toxic LO2 cells. Moreover, prevented oxidative damage induced by H2O2. Conclusion These findings highlight therapeutic potential treatment, warranting further clinical investigation.

This research endeavored to elucidate the antioxidant attributes of lactic acid bacteria, specifically their impact on anti-aging and lifespan augmentation in Caenorhabditis elegans. The study focused Lactiplantibacillus plantarum A72, identified through ARTP mutagenesis for its potent properties. In vitro analysis affirmed free radical neutralizing capacity. C. elegans, strain not only extended by 25.13% amplified motility 2.52-fold, but also maintained reproductive capabilities. Remarkably, Lpb. A72 diminished reactive oxygen species (ROS) malondialdehyde (MDA) levels elegans 34.86% 69.52%, respectively, while concurrently enhancing enzyme activities. bolstered survival rates 46.33% 57.78% under high temperature H2O2 conditions, respectively. Transcriptomic scrutiny revealed that could retard aging extend upregulating sod-5 hsp-16.1 genes downregulating fat-6 lips-17 genes. These findings propose as a potential bacteria.

Lactic acid bacteria (LAB), a probiotic, provide various health benefits. We recently isolated new Lactobacillus paracasei strain with strong anti-inflammatory effects under lipopolysaccharide-induced conditions and proposed mode of action—augmenting the endoplasmic reticulum stress pathway for functions in host cells. The beneficial L. strains on skin have been described; however, paracasei-derived extracellular vesicles (LpEVs) are poorly understood. Herein, we investigated whether LpEVs can improve inflammation-mediated phenotypes by determining their primary human cells three-dimensional (3D) full-thickness equivalent tumor necrosis factor (TNF)-α-challenged inflammatory conditions. were efficiently taken up much less cytotoxic to than bacterial lysates. Furthermore, low LpEV concentrations restored TNF-α-induced cellular phenotypes, resulting increased cell proliferation collagen synthesis, but decreased levels (matrix metalloproteinase 1, interleukin 6, 8) dermal fibroblasts, which was comparable that retinoic acid, representative antiaging compound. validated 3D model. treatment remarkably epidermal malformation, abnormal keratinocytes basal layer, reduction synthesis. Additionally, penetrated reached deepest layer within 24 h when overlaid top equivalent. they possessed superior antioxidant capacity compared cell-derived EVs. Taken together, probiotic be attractive substances improving inflammation-induced disorders.
