The aetiologies and origins of neurodegenerative diseases, such as Alzheimer's disease (AD), Parkinson's (PD), amyotrophic lateral sclerosis (ALS) Huntington's (HD), are complex multifaceted. A growing body evidence suggests that the gut microbiome plays crucial roles in development progression diseases. Clinicians have come to realize therapeutics targeting potential halt This narrative review examines alterations AD, PD, ALS HD, highlighting close relationship between brain Processes mediate microbiome-brain communication including immunological, vagus nerve circulatory pathways, evaluated. Furthermore, we summarize for diseases modify its metabolites, diets, probiotics prebiotics, microbial antibacterials faecal transplantation. Finally, current challenges future directions discussed.

Abstract Purpose of Review This review explores the intricate relationships among gut microbiota, dietary patterns, and mental health, focusing specifically on depression. It synthesizes insights from microbiological, nutritional, neuroscientific perspectives to understand how gut-brain axis influences mood cognitive function. Recent Findings studies underscore central role microbiota in modulating neurological psychological health via axis. Key findings highlight importance components, including probiotics, prebiotics, psychobiotics, restoring microbial balance enhancing regulation. Different patterns exhibit a profound impact composition, suggesting their potential as complementary strategies for support. Furthermore, mechanisms like tryptophan metabolism, HPA axis, metabolites such SCFAs are implicated linking diet Clinical trials show promising effects probiotics alleviating depressive symptoms. Summary illuminates diet-based interventions targeting mitigate depression improve health. While interplay between diversity, diet, brain function offers therapeutic avenues, further clinical research is needed validate these establish robust, individualized treatment strategies.

ABSTRACT Advancing microbiome–gut–brain axis science requires systematic, rational and translational approaches to bridge the critical knowledge gaps currently preventing full exploitation of gut microbiome as a tractable therapeutic target for gastrointestinal, mental brain health. Current research is still marked by many open questions that undermine widespread application humans. For example, lack mechanistic understanding probiotic effects means it remains unclear why even apparently closely related strains exhibit different in vivo. live microbial psychobiotics, consensus on their adjunct treatments conventional neuromodulators, use unmedicated populations or at‐risk cohorts with sub‐clinical symptomatology warranted. Electroacupuncture (EA) has shown promise as a treatment for Functional constipation (FC), with growing evidence suggesting it may enhance gut motility. MicroRNAs (miRNAs) serve key regulatory molecules mediating host-microbiota interactions. However, the specific fecal miRNAs regulating microbiota composition and metabolism in EA-treated constipated mice, along their targets, remain unidentified. We examined microbiome composition, metabolism, colonic miRNA expression loperamide-induced mice to identify differentially expressed assess relationships microbial abundance, An antibiotic cocktail adeno-associated virus were employed interfere target vivo, thereby validating proposed mechanism. Our results indicate that miR-205-5p, significantly upregulated tissues of promotes intestinal motility microbiome-dependent manner. Specifically, EA promoted growth Lactobacillus reuteri , enriched feces constipation-recovered through host-derived miR-205-5p regulation. Furthermore, its tryptophan metabolites (indole-3-acetamide, indole-3-acetic acid, indole-3-carboxaldehyde) alleviated constipation. These findings underscore pivotal role modulating EA.
